7.1. ÖZET Fare deri karsinogenez modelinde Amifostin'in kemopreventif etkisi var mı ? Amifostine, kemoterapi ve radyoterapiye bağlı toksisitelere karşı normal dokular için selektif bir sitoprotektif ajandır. Bu çalışmada, fare deri karsinogenez modelinde amifostinin muhtemel kemopreventif etkileri araştırılmıştır. Amifostine için tek ve tekrarlanan dozların MTD değerleri sırasıyla, 400 mg/kg ve 200 mg/kg bw olarak bulunmuştur. CD-1 farelere başlangıçta inisiasyon amaçlı 100 mmol DMBA uygulanmış ve daha sonra promosyon amaçlı 10 nmol TPA ile, 22 hafta boyunca iki haftada bir olmak üzere devam edilmiştir. Farelerin sırtlarına 40 mg/kg bw' lik amifostine dozu farelerin tıraşlanmış sırtlarına ilk grupta DMBA uygulanmasından önce.ikinci grupta her TPA uygulamasından önce ve üçüncü grupta ise hem DMBA hem de TPA uygulamalarından önce subkutan olanrak uygulanmıştır. Her faredeki tümör sayısı ve tümörlü fare sayısı peryodik olarak değerlendirilmiştir. Yirmiikinci haftada tümör multiplisitesi ve tümör insidansı sırasıyla kontrol grubunda, 20,13 ve 95,80, birinci grupta 20,28 ve100, ikinci grupta 30,05 ve 100, üçüncü grupta ise 30,46 ve 96,10 olarak bulunmuştur. Hernekadar 22. haftadaki sonuçlar istatistiksel olarak anlamlı olmasa da üçüncü grupta, papillomların gelişiminde dört haftalık bir gecikme kaydedilmiştir.

7.2. SUMMARY Does amifostine have any chemopreventive effect in mouse skin tumorigenesis? Amifostine (WR-2721), is a selective cytoprotective agent for normal tissues from the toxicities associated with chemotherapy and irradiation. In this study, the possible chemopreventive effects of amifostine was searched in mouse skin tumorigenesis.The MTD values of amifostine was searched in single and repetetive administrations were determined as 400 mg/kg bw dose and 200 mg/kg bw respectively. CD-1 mice were initiated by 1 00 mmol DMBA and promoted by 1 0 nmol TPA twice a week for 22 weeks. A 40 mg/kg bw dose of amifostine was administered to the shaved back of mice subcutaneously before DMBA in the first group and before each TPA in the second group and before both agents in the third group. The number of tumors in each mouse and the number of mice with tumors were determined periodically. The tumor multiplicity and the tumor incidence in 22.nd week were found as 20.13 and 95.80 in the control group, 20.28 and 100 in the first group, 30.05 and 100 in the second group, 30.46 and 96.10 in the third group respectively. Although the results in 22.nd week were not statistically significant, a delaying effect of amifostine on the genesis of papillomas for four weeks noted in the third group was significant.
