Do noxious events always induce aversive memories? Contrary to popular belief, noxious events do not exclusively induce aversive memories. A cue that predicts a punishment will cause an aversive memory for this cue. However, if the cue is presented shortly after the cessation of an unconditioned punishing cue, it induces an appetitive memory. This pairing between the cue and the termination of aversive event is known as relief learning. Compared to the other kinds of Pavlovian learning such as fear learning or reward learning, relief learning is poorly understood. Previous studies have revealed a critical role of nucleus accumbens in conditioned relief memory. In this study, I investigated the neuropharmacology of relief learning. More specifically, I aimed to understand if dopaminergic transmission within the nucleus accumbens modulated the acquisition of relief learning. If so, which dopamine receptor subtype is mainly involved? To address these questions, specific dopamine receptor 1 or dopamine receptor 2 antagonists were locally injected into the nucleus accumbens of Sprague-Dawley rats, and the alterations in relief learning were examined by the relief-induced modulation of the startle response. My results clearly demonstrate the involvement of dopaminergic transmission within the nucleus accumbens in the acquisition of conditioned relief memories, and both D1 and D2 receptors appear to critically contribute to this process. These findings might shed light on the etiology of some odd behaviors and anxiety disorders.
