Bifidobacteria commonly used for fermented milk. The level of bifidobacteria in fermented milk should be 106 CFU/ml. Eleven probiotic yogurt samples were tested and these were purchased from UK domestic markets and stored 4 °C until the end of shelf-life. Many culture media have been developed to enumerate and isolate bifidobacteria even though bifidobacteria were enumerated and isolated using selective TOS-MUP agar modified by the addition of mupirocin (50 μg/ml). Bifidobacteria has been resisted for the mupirocin antibiotic so that it has been inhibited non-bifidobacteria on agar. Gram staining, catalase test and oxidase rapid tests were performed for the phenotypic identification of bifidobacteria. Genotypic identification was carried out by PCR method using 16S rRNA and species-specific primers. Sequences were present in GenBank database. For the acid and bile tolerance of bifidobacteria was used TOS agar. To realize antimicrobial susceptibility of bifidobacteria was used TSB agar containing 0.8% w/v agar and two different methods namely normal spot test and well-diffusion test were performed. The purpose of this study was to enumerate and isolate bifidobacteria in probiotic yogurts and identify bifidobacteria phenotypic and genotypic characteristic as well as including Gram staining, catalase, oxidase tests and PCR method. In addition, this study was carried out to examine probiotic characterisation of bifidobacteria besides acid, bile and antimicrobial tests were performed.
