Özet

Bu tez çalışmasında, yara iyileşmesini desteklemek amacıyla PVA (polivinil alkol) ve PVP (polivinil pirolidon) esaslı hidrojel yara örtüleri geliştirilmiş; bu hidrojellere doğal biyoaktif özellik kazandırmak için Alpinia galanga, Salvadora persica ve Corchorus olitorius bitki ekstraktları ile ayrıca bu üç ekstraktın karışımı ayrı formülasyonlar halinde sisteme dahil edilmiştir. Yara iyileşmesi hemostaz, inflamasyon, proliferasyon ve yeniden şekillenme fazlarını içeren çok basamaklı bir süreç olup; enfeksiyon, aşırı kuruma ve oksidatif stres gibi etkenler iyileşmeyi yavaşlatabilmektedir. Bu nedenle modern yara örtülerinden beklenen; uygun nem dengesini korumak, eksüdayı yönetmek, dokuya uyumlu olmak, yaraya yapışmayı azaltmak ve mümkünse antibakteriyel/antioksidan destek sağlayarak iyileşme mikro çevresini iyileştirmektir. Hidrojeller yüksek su içeriği, yumuşak dokuya benzer yapıları ve aktif ajan taşıma potansiyelleri nedeniyle bu amaç için güçlü adaylardır. Hidrojel matrisi, kimyasal çapraz bağlayıcı kullanılmadan dondurma–çözündürme (freeze–thaw) yöntemiyle fiziksel olarak çapraz bağlanmış şekilde hazırlanmıştır. PVA ve PVP belirlenen oranlarda sulu ortamda çözündürülerek homojen polimer karışımı elde edilmiş, kalıplara dökülmüş ve ardışık dondurma–çözündürme döngüleriyle üç boyutlu hidrofilik ağ yapı oluşturulmuştur. Bitki ekstraktları uygun çözücü sistemiyle hazırlanıp saklanmış; hidrojellere yükleme aşamasında iki farklı strateji karşılaştırılmıştır: üretim sırasında (ÜSY) yükleme yaklaşımında ekstraktlar jel oluşumundan önce polimer çözeltisine eklenerek döngüler ekstrakt varlığında yürütülmüş; sonradan yükleme (SY) yaklaşımında ise önce ekstraktsız (boş) hidrojel hazırlanıp kurutulmuş/liyofilize edilmiş ve daha sonra ekstrakt çözeltisi hidrojellere emdirilerek yükleme gerçekleştirilmiştir. Böylece ekstraktın ağ yapı içindeki dağılımı, matrise bağlanma/entegrasyon düzeyi ve yüzeyde erişilebilirliği gibi parametrelerin, hidrojel performansına etkisi sistematik biçimde değerlendirilmiştir. Üretilen örneklerde FTIR analizi ile PVA/PVP'ye ait karakteristik bantların korunduğu, ekstrakt ilavesinin polimer omurgasını bozmadığı ve özellikle ÜSY yüklemede polimer–ekstrakt etkileşimlerinin (hidrojen bağları vb.) daha belirgin hale gelebildiği ortaya konmuştur. Şişme testi ve su tutma değerlendirmeleri, tüm numunelerin hidrofilik karakter gösterdiğini ve kısa sürede su alarak dengeye yaklaştığını göstermiştir. Şişme davranışı açısından SY grubunda genel olarak daha yüksek değerler elde edilirken, su tutma performansının ekstrakt türüne bağlı olarak değiştiği belirlenmiştir. Nitekim AG ve Karışım formülasyonlarında SY yöntemi daha yüksek su tutma kapasitesi sağlarken, SP ve CO içeren formülasyonlarda ÜSY yöntemi daha yüksek kalan kütle değerleri ile daha avantajlı bir profil ortaya koymuştur. Jel fraksiyonu (jelleşme derecesi) sonuçları, örneklerin yapısal bütünlüğünün yükleme stratejisine ve ekstrakt tipine bağlı olarak değişebildiğini; bazı durumlarda sonradan yükleme (SY) yaklaşımının ağ stabilitesini daha avantajlı seviyede tutabildiğini desteklemiştir. Temas açısı ölçümleri ise tüm numunelerde 90°'nin altında değerlerle hidrofilik yüzey davranışının sürdüğünü ve bitki ekstraktlarının yüzey ıslanabilirliğini artırabildiğini göstermiştir; bu durum yara yüzeyiyle etkileşim ve sıvı temasının kolaylaşması açısından olumlu kabul edilmiştir. Taramalı elektron mikroskobu (SEM) görüntüleri, yükleme stratejisinin hidrojelin yüzey morfolojisi ve gözenekli yapısı üzerinde belirgin bir etkiye sahip olduğunu göstermiştir. Boş PVA/PVP hidrojeli genel olarak sürekli ve nispeten homojen bir morfoloji sergilerken, ekstrakt içeren numunelerde yöntem bağımlı olarak artan pürüzlülük, mikro-domen oluşumu ve yer yer mikro/sub-mikron düzeyde porozite gözlenmiştir. Üretim sırasında yükleme (ÜSY) uygulanan örneklerde ekstraktın ağ oluşumu esnasında matrise entegre olmasıyla daha bütünleşik bir doku ve daha düzenli dağılım izlenirken; sonradan yükleme (SY) uygulanan örneklerde ekstraktın yüzey ve erişilebilir gözenek bölgelerinde yoğunlaşmasına bağlı olarak daha heterojen bir görünüm, lokal aglomerasyonlar ve yüzeye yakın partiküler birikimler dikkat çekmiştir. SEM bulguları, ekstraktın matristeki konumlanmasının ıslanabilirlik, şişme ve salım davranışı gibi performans parametrelerini dolaylı olarak yönlendirebileceğini desteklemektedir. Antibakteriyel etkinlik agar difüzyon yöntemiyle E. coli ve S. aureus'a karşı değerlendirilmiş; boş PVA/PVP hidrojelin inhibisyon zonu oluşturmaması matriksin tek başına antibakteriyel olmadığını göstermiştir. Bitki ekstraktı yüklü hidrojellerde ise inhibisyon zonlarının oluşması, biyoaktif bileşenlerin ortama difüze olarak bakteriyel büyümeyi baskılayabildiğini ortaya koymuştur. DPPH testi sonuçları da benzer biçimde boş hidrojelin düşük/ihmal edilebilir antioksidan etki sergilediğini, ekstrakt yüklü örneklerde ise serbest radikal süpürme kapasitesinin belirgin şekilde arttığını göstermiştir. Genel olarak, yükleme stratejisi ekstraktın hidrojel içindeki konumu ve salım dinamiklerini etkileyerek antibakteriyel ve antioksidan performansı belirleyebilmektedir. İlaç taşıyıcı potansiyelini doğrulamak amacıyla model antibiyotik olarak gentamisin kullanılarak in vitro ilaç salımı değerlendirilmiştir. Eşit kuru numune kütlesi (55 mg) kullanılarak ÜSY ve SY için benzer yükleme miktarları sağlanmış (ÜSY: 573 µg; SY: 553 µg) ve buna karşılık gelen ilaç yükleme kapasiteleri sırasıyla yaklaşık %1,04 ve %1,01 olarak hesaplanmıştır. Salım kinetiği, yükleme stratejisinin özellikle başlangıç evresini belirgin biçimde etkilediğini göstermiştir: SY örneklerinde 0,5 saatte %25,00 (137,64 µg) ile daha belirgin bir burst salım gözlenirken, ÜSY örneklerinde aynı sürede %17,10 (97,97 µg) salım gerçekleşmiştir. 12. saatte kümülatif salım SY için %56,20 (309,42 µg), ÜSY için %43,21 (247,59 µg) düzeyinde bulunmuştur. Bu farkın, SY yaklaşımında ilacın daha çok yüzey/yüzeye yakın gözenek bölgelerinde konumlanmasıyla kısa difüzyon yolu ve hızlı başlangıç salımının desteklenmesine; ÜSY yaklaşımında ise ilacın ağ yapısına daha derin entegrasyonu nedeniyle daha kontrollü bir salım profiline bağlanabileceği değerlendirilmiştir.

This thesis focuses on the development of polymeric hydrogel wound dressings designed to support wound healing by providing a moist microenvironment and by introducing additional biofunctional performance through natural plant extracts. Wound healing is a complex, multi-phase biological process (hemostasis, inflammation, proliferation, and remodeling) that can be delayed or complicated by infection, excessive dehydration, and oxidative stress. Therefore, an ideal wound dressing is expected to protect the wound surface, manage exudate, preserve moisture balance, reduce microbial burden, and contribute to biochemical support such as antioxidant protection. In this context, hydrogels are widely investigated as modern wound dressing candidates because of their high-water content, soft-tissue compatibility, and ability to serve as carriers for therapeutic agents. Within the scope of the study, PVA (polyvinyl alcohol) and PVP (polyvinylpyrrolidone) were selected as the base polymers due to their established biocompatibility and suitability for forming hydrophilic networks. The thesis builds the experimental design around a key methodological comparison: incorporating bioactive agents into PVA/PVP hydrogels via two different strategies—in-situ loading (adding the extract during hydrogel formation) versus post-loading (loading the extract into a pre-formed hydrogel). The plant extracts investigated are Alpinia galanga (galangal), Salvadora persica (miswak), Corchorus olitorius (molokhia/ molehiya), and also a mixture of these three extracts which were considered as natural sources of compounds with potential antibacterial and antioxidant relevance for wound care. In addition, a pharmaceutical antibacterial reference (Augmentin) is used as a control for benchmarking antibacterial performance. The overarching aim is to determine whether the hydrogels can be formed reliably, how extract incorporation changes physicochemical behavior, and which loading strategy provides more effective functional outcomes—especially antibacterial activity against E. coli and S. aureus, and antioxidant activity in the DPPH assay. Experimentally, the hydrogel matrix is prepared using a physical crosslinking approach based on the freeze–thaw (freezing–thawing) method, avoiding chemical crosslinkers and thereby reducing concerns about residual toxic reagents. In a typical workflow, PVA and PVP are dissolved to form a homogeneous polymer mixture and then cast into molds (Petri dishes) prior to repeated freeze–thaw cycles. During freeze–thawing, crystalline micro-domains and polymer–polymer interactions act as physical crosslink points, generating a stable hydrogel network. After formation, hydrogels are shaped into standardized samples and lyophilized (freeze-dried) to enable consistent downstream testing and to facilitate controlled post-loading. For in-situ loading, each plant extract is introduced into the polymer solution before gelation, using a defined loading ratio (reported in the thesis as 0.35 g extract per 1 g polymer, weighed accordingly and mixed prior to casting). For post-loading, blank hydrogels are first formed and dried; then loading is achieved by placing the hydrogel pieces in an extract solution volume matched to the absorption capacity of the samples, typically under agitation (e.g., orbital shaking) until the solution is taken up, after which samples are dried again under controlled conditions. A critical experimental intention is that the mass of loaded extract in post-loading is selected to be comparable to the in-situ target, enabling a fair comparison between strategies. To evaluate how these design choices affect structure and performance, the thesis applies a set of physicochemical characterization tests commonly used for hydrogel wound dressings. FTIR spectroscopy is used to confirm the preservation of the polymer backbone and to assess whether extract incorporation produces detectable changes in the spectral profile. The reported FTIR overlays indicate that the characteristic PVA/PVP bands remain present across samples, including the broad O–H stretching band (around the ~3300 cm⁻¹ region), C–H stretching (near ~2900 cm⁻¹), and carbonyl-related signals associated with PVP (around ~1650 cm⁻¹), along with fingerprint-region signals consistent with the polymer network. The main interpretation is that extract loading—whether in-situ or post-loading—does not destroy the fundamental chemical identity of the PVA/PVP matrix; instead, the extract is incorporated into or onto the network primarily through physical interactions (e.g., hydrogen bonding and entrapment in the porous structure), which is consistent with the choice of physical crosslinking. The moisture-handling behavior is evaluated through swelling and water retention tests, because these properties strongly influence wound dressing performance: swelling relates to exudate uptake and fluid buffering, while retention reflects how well the hydrogel can maintain hydration over time. In the swelling experiments, the thesis tracks mass changes after immersing dried hydrogels in distilled water and calculates swelling ratio as a function of time using the standard gravimetric approach (W₀ and Wₜ). A consistent pattern emerges: swelling rises rapidly at early time points and then approaches a plateau, reflecting diffusion-controlled water uptake until equilibrium. Importantly, adding extracts alters swelling behavior relative to the blank PVA/PVP hydrogel. In-situ incorporation can reduce swelling compared with the blank, which is discussed as a consequence of extract components occupying free volume, narrowing diffusion pathways, and potentially increasing effective network density. In post-loading, swelling behavior may remain closer to the blank or differ depending on extract type, because the extract is primarily introduced after the network is already formed; thus, the base network formation is less disrupted, and swelling becomes more strongly linked to how the extract distributes within pores and at pore walls rather than how it interferes with gelation. The water retention test further clarified that retention behavior depended on both the extract type and the loading strategy. In all groups, hydrated samples gradually lost mass over time, indicating progressive water release from the network; however, the remaining mass values at later time points differed among formulations. Post-loading provided higher water retention in AG and mixture-containing hydrogels, whereas in-situ loading resulted in higher remaining mass values in SP- and CO-containing formulations. These findings indicate that water retention cannot be generalized in favor of a single loading strategy, but should instead be interpreted according to the specific extract–matrix interaction in each formulation Network stability is further addressed through the gel fraction / gelation degree approach described in the thesis. After swelling and subsequent controlled drying, the remaining stable mass is used to quantify the extent to which the hydrogel network resists dissolution and maintains its integrity. The central conclusion drawn is that the freeze–thaw method provides a sufficiently stable physical network for handling and testing, while extract incorporation can modulate (but not eliminate) network stability. This matters practically because wound dressings must remain intact during application, hydration, and removal, rather than dissolving or fragmenting prematurely. Surface wettability is characterized using contact angle measurements by depositing a small water droplet (reported as 4 µL) onto the sample surface. The thesis interprets contact angle values in the standard way: angles below 90° indicate hydrophilic behavior, which is generally preferred for wound dressings because it supports wetting and fluid interaction. Across the sample set, the hydrogels are discussed as maintaining hydrophilic character, consistent with the chemistry of PVA/PVP and the water-friendly nature of the polymer network. Differences between samples are discussed as being linked to surface composition and microstructure (including how extracts distribute at the surface). In principle, post-loading can strengthen surface-localized effects because the extract is introduced through absorption into the already-formed network, making it more likely that a portion remains close to pore walls and surfaces. In-situ loading can distribute the extract more throughout the bulk, potentially reducing how strongly surface chemistry changes relative to the blank, depending on the extract–polymer interactions. Beyond physicochemical behavior, the thesis emphasizes biological performance, especially antibacterial activity and antioxidant capacity, because these are directly connected to wound healing outcomes. Antibacterial evaluation is performed against two clinically relevant bacteria: E. coli (Gram-negative) and S. aureus (Gram-positive). The discussion recognizes that Gram-negative bacteria often present higher resistance in diffusion-based assays due to the protective outer membrane, which can reduce sensitivity to certain agents. In the antibacterial test outcomes, the control antibiotic (Augmentin) shows the expected strong inhibitory effect, validating the test conditions and confirming that the assay can clearly detect meaningful antibacterial performance when potent antibacterial agents are present. Plant-extract hydrogels, by contrast, display weak to moderate inhibition depending on extract type and loading method. A consistent interpretation is that antibacterial effectiveness is generally more pronounced against S. aureus than against E. coli, aligning with the structural differences between Gram-positive and Gram-negative bacteria and with the idea that some plant-derived compounds may diffuse and act more effectively in the Gram-positive context. Crucially, the thesis uses these antibacterial outcomes to highlight the functional difference between loading strategies. Post-loading often produces clearer inhibition behavior in diffusion-based measurements because active compounds are more readily available near the surface and can migrate into the surrounding medium more rapidly. This surface availability supports stronger early release and thus can yield more visible zones of inhibition. In-situ loading, while beneficial for distributing extract through the matrix, may trap a larger fraction of bioactives within the hydrogel bulk, which can reduce immediate diffusion into the agar and weaken apparent inhibition in short-term plate assays—even if the system could potentially support a more sustained release profile under longer or different conditions. Therefore, the method comparison is not treated as a simple“better versus worse”decision; rather, it is framed as a trade-off between rapid surface availability (post-loading) and deeper matrix integration (in-situ), with implications for how the dressing might perform depending on wound needs (acute bacterial challenge versus longer-term controlled delivery). The antioxidant evaluation is performed using the DPPH radical scavenging assay, a standard chemical method for estimating the ability of a sample to neutralize free radicals. The thesis reports that extract-functionalized hydrogels demonstrate meaningful radical scavenging behavior, whereas the blank PVA/PVP hydrogel—expectedly—shows negligible antioxidant activity. This result supports the conclusion that antioxidant components from the plant extracts remain active after incorporation and that the hydrogel can serve as a carrier that preserves functional antioxidant potential. From a wound healing perspective, this is important because oxidative stress is a known factor that can impair healing progression; thus, providing local antioxidant functionality may contribute to a microenvironment more favorable for tissue recovery. As with antibacterial activity, the loading method matters because it influences how readily antioxidant compounds can interact with the surrounding medium: post-loading can enhance near-surface availability for rapid interaction, while in-situ loading can embed antioxidants more deeply, potentially supporting longer-lasting antioxidant release rather than immediate maximal effect. Taken together, the thesis concludes that PVA/PVP hydrogels produced by freeze–thaw cycling form stable, hydrophilic wound dressing candidates with tunable moisture management properties. Incorporation of plant extracts (galangal, miswak, molokhia) successfully adds biofunctional performance, particularly antioxidant capacity and measurable antibacterial activity, though plant-based antibacterial effects remain lower than the pharmaceutical antibiotic reference. Importantly, the comparative analysis shows that loading strategy is a decisive parameter: in-situ loading more strongly couples the extract to the network formation process and may reduce swelling or water uptake when the extract limits diffusion space, whereas post-loading typically preserves base network formation and tends to improve the immediate availability of active compounds at surfaces and pores, which is advantageous for diffusion-dependent antibacterial performance and rapid chemical interaction in assays such as DPPH. In final evaluation, the developed dressings are presented as promising multifunctional platforms, especially when the intended goal is to combine moist wound coverage with natural bioactivity. The thesis highlights that the optimal choice between in-situ and post-loading should be made based on the targeted therapeutic profile: post-loading is favored when rapid, surface-accessible activity is needed, while in-situ loading is meaningful when deeper incorporation and potential sustained release are desirable, even if initial diffusion-based inhibition appears weaker. Overall, the work supports the feasibility of producing plant-extract functionalized PVA/PVP hydrogel wound dressings using a physically crosslinked approach and demonstrates that the method of loading is not a minor technical detail but a central design factor controlling the dressing's physicochemical and biofunctional performance.
