Özet

Polilaktik asit (PLA), kolay işlenebilirliği, düşük üretim maliyeti, biyouyumluluğu ve biyobozunur yapısı sayesinde gıda ambalajı uygulamaları için umut vadeden bir poliesterdir. Ancak kırılgan yapısı, düşük termal direnci, yavaş kristalizasyonu, düşük kopma uzaması ile zayıf su buharı ve oksijen bariyeri gibi dezavantajları kullanımını sınırlandırmaktadır. Bu tez çalışmasında, PLA'nın bu sınırlılıklarını azaltmak amacıyla nanokristal selüloz (NKS) ve lignin (L) kullanılarak çözelti dökme yöntemiyle; tek katmanlı kontrol PLA, PLA–NKS, PLA–L ve PLA–NKS–L filmleri ile birinci katmanı PLA–NKS, ikinci katmanı lignin (PLA–NKS/L) ve lignin–NKS (PLA–NKS/L–NKS) olan iki katmanlı kompozit filmler geliştirilmiştir. Tek katmanlı filmlerin üretiminde, PLA matrisi plastikleştirici olarak ağırlıkça %5 oranında polietilen glikol (PEG 400) içerecek şekilde kloroform içerisinde çözülmüş; homojen bir film yapısı elde edebilmek amacıyla çözeltiye ardışık manyetik karıştırma ve ultrasonikasyon işlemleri uygulanmıştır. PLA–NKS filmlerinde ağırlıkça %2 oranında NKS, PLA–L filmlerinde ağırlıkça %2 oranında lignin kullanılmış; PLA–NKS–L filmlerinde ise NKS ve lignin birlikte ağırlıkça %2 oranında ilave edilmiştir. Hazırlanan film çözeltileri petri kaplarına dökülmüş ve 25 ± 2 °C sıcaklık ile %50 ± 5 bağıl nemde 24 saat boyunca kurutularak filmler elde edilmiştir. İki katmanlı filmlerin üretiminde birinci katman, PLA–NKS esaslı olarak hazırlanmış; ikinci katmanlar ise lignin ve lignin–NKS'nin 0,01 M sodyum hidroksit çözeltisi kullanılarak uzun süreli karıştırma ve ultrasonikasyon işlemleri sonrasında elde edilmiştir. İki katmanlı filmlerin üretiminde, birinci katman 25 ± 2 °C sıcaklık ve %50 ± 5 bağıl nem koşullarında 18 saat süreyle ön kurutmaya tabi tutulmuş; ikinci katmanların dökülmesini takiben, filmler aynı sıcaklık ve bağıl nem koşullarında 24 saat boyunca kurutularak elde edilmiştir. Geliştirilen tek ve iki katmanlı kompozit filmler; fiziksel (kalınlık, renk, çözünürlük, şişme indeksi ve temas açısı), yapısal ve morfolojik (FTIR, XRD ve FE-SEM), termal (DSC), mekanik (çekme mukavemeti, kopma anında uzama ve ısıl yapışma mukavemeti), bariyer (su buharı geçirgenliği, ışık geçirgenliği ve opaklık) ve aktif (antioksidan ve antimikrobiyal) özellikler açısından kapsamlı olarak değerlendirilmiştir. Filmlerin kalınlığı 31–56 µm aralığında olup, NKS ve/veya lignin ilavesi kalınlığı anlamlı şekilde artırmıştır (p<0,05). Optik performansta lignin ve katmanlı tasarım belirleyici olmuş; kontrol PLA en yüksek parlaklığı (L* = 94,54) gösterirken, iki katmanlı PLA–NKS/L–NKS filmi en düşük parlaklığı (L* = 55,28) ve en yüksek renk değişimini (ΔE = 54,66) sergilemiştir. Bu durum, lignin yapısındaki kromofor gruplarının ışık absorbsiyonunu artırmasıyla ilişkilendirilmiştir. Özellikle iki katmanlı filmlerde ligninin yüzeyde yüksek oranda bulunması, tek katmanlı filmlerde gözlenmeyen belirgin kahverengimsi ton oluşumuna neden olmuş; bu durum optik özelliklerin yanı sıra UV ışınımının etkin biçimde soğurulmasını sağlayarak filmlerin UV bariyer performansının güçlenmesinde kritik bir rol oynamıştır. Tek katmanlı filmler arasında çözünürlük ve şişme indeksi açısından fark görülmezken (p>0,05), iki katmanlı PLA–NKS/L ve özellikle PLA–NKS/L–NKS grubunda çözünürlük (%6,72) ve şişme indeksi (%1,39) en yüksek değerlere ulaşmıştır (p<0,05). Temas açısı sonuçları da bu bulguları destekler nitelikte olup, kontrol PLA'da 0. dakikada 68,5° olarak belirlenen temas açısı, iki katmanlı filmlerde L yüzeyinde 56,3° ve L–NKS yüzeyinde 54,7° olarak ölçülmüştür. L ve L–NKS katmanlarının FTIR spektrumlarında hidroksil gruplarına (–OH) ait titreşimlerin korunması ve aromatik/fenolik piklerin varlığı, polar grupların erişilebilirliğinin artmasına bağlı olarak su ile etkileşimin güçlendiğini desteklemiştir. Yapısal analizlerden biri olan XRD'de, kontrol PLA'nın tipik kırınımları 2θ = 14,9°, 16,9° ve 18,5°'de gözlenmiştir. PLA–NKS filminde 22,4° civarındaki pik NKS varlığını doğrularken, PLA–L ve PLA–NKS–L filmlerinde pik şiddetlerindeki azalma ligninin amorf yapısının kristal fazı kısmen baskılayabileceğini düşündürmüştür. İki katmanlı PLA–NKS/L–NKS filminde 22,8°'deki selüloza özgü pikin belirginleşmesi, bu yapıda selülozun kristal fazının daha belirgin hâle geldiğini göstermiştir. FE-SEM analizlerinde kontrol PLA yüzeyi düzgün ve homojen bir yapı sergilerken; PLA–NKS filmlerinde pürüzlülükte hafif artış, PLA–L filmlerinde mikro boşluklar ve PLA–NKS–L filmlerinde yer yer kümelenmiş yapılar gözlenmiştir. İki katmanlı filmlerde L ve L–NKS katmanları daha pürüzlü, lifsi ve iç içe geçmiş yapılar sergilerken; PLA–NKS katmanı daha düzgün ve sürekli bir morfoloji göstermiş, böylece katmanlı yapının oluşumu doğrulanmıştır. Termal özellikler incelendiğinde, camsı geçiş sıcaklığının tüm film gruplarında benzer değerler (58,9–59,0 °C) sergilediği tespit edilmiştir. Tek katmanlı filmlerde erime sıcaklığı 155,6–155,8 °C aralığında değişmezken, iki katmanlı filmlerde hafif bir artış eğilimi görülmüştür. PLA–NKS filmlerinde soğuk kristalizasyon sıcaklığındaki düşüş, NKS ilavesine bağlı çekirdeklenme etkisini; PLA–L ve PLA–NKS–L filmlerindeki artış ise lignin ilavesinin zincir hareketliliğini kısmen sınırladığını göstermiştir. Genel olarak, DSC sonuçları katkı maddelerinin PLA'nın camsı geçiş sıcaklığını değiştirmediğini, erime sıcaklığı üzerinde kısıtlı bir etki gösterdiğini ve esas etkinin soğuk kristalizasyon sıcaklığı üzerinde yoğunlaştığını ortaya koymaktadır. Mekanik açıdan PLA–NKS filminde, kontrol PLA'ya (33,03 MPa; %2,11) kıyasla çekme mukavemeti 40,54 MPa'ya yükselmiş ve kopma anında uzama %1,73'e düşmüştür. İki katmanlı PLA–NKS/L filmleri, en düşük çekme mukavemeti (28,86 MPa) ve en yüksek kopma anında uzama (%2,69) değerlerini gösterirken; PLA–NKS/L–NKS filmleri, daha yüksek çekme mukavemeti (36,26 MPa) ve görece yüksek kopma anında uzama (%2,32) değerleri göstermiştir. Tüm filmler ısıl olarak yapıştırılabilir bulunmuş; en düşük ısıl yapışma PLA–NKS/L'de (165,98 N/m), en yüksek değer ise tek katmanlı PLA–NKS–L'de (315,44 N/m) ölçülmüştür. Bariyer özellikler açısından değerlendirildiğinde, en düşük su buharı iletim hızı tek katmanlı PLA–NKS filminde (5,08 g/m².24 saat), en yüksek değer iki katmanlı PLA–NKS/L filminde (7,40 g/m².24 saat) elde edilmiştir. UV bariyer performansı açısından PLA–NKS/L–NKS filmi UV-C'de %100, UV-B'de %99,8, UV-A'da %98,8 ve görünür bölgede %81,8 azalma sağlayarak en güçlü UV ve ışık bariyeri özelliğini göstermiştir. Lignin içermeyen filmlerde etkin bir antioksidan aktivite gözlenmemiş; lignin içeren tek katmanlı filmlerde %40–50, iki katmanlı filmlerde ise %85'e ulaşan antioksidan aktivite elde edilmiştir. Tek katmanlı filmlerde antibakteriyel veya antifungal aktivite gözlenmemiştir. Buna karşılık, iki katmanlı filmlerde Escherichia coli, Staphylococcus aureus, Salmonella typhimurium ve Bacillus subtilis'e karşı 1,1–7,9 mm aralığında inhibisyon zonları oluşmuş; ayrıca Aspergillus niger'e karşı antifungal aktivite belirlenmiştir. Bu bulgular, antimikrobiyal etkinliğin yalnızca bileşime değil, katmanlı tasarım, aktif bileşenlerin miktarı ve yüzeydeki erişilebilirliğine güçlü biçimde bağlı olduğunu göstermektedir. Elde edilen bulgular, PLA bazlı kompozit filmlerde NKS ilavesinin ağırlıklı olarak mekanik performans üzerinde; lignin ilavesinin ise özellikle optik özellikler ile antioksidan ve antimikrobiyal aktivite üzerinde belirleyici bir rol oynadığını göstermiştir. Katmanlı film tasarımı aktif bileşenlerin yüzeydeki erişilebilirliğini artırarak UV bariyer özelliği, antioksidan ve antimikrobiyal etkinin geliştirilmesine olanak sağlamış, yüzey özelliklerinin katmanlı yapı aracılığıyla yönlendirilebileceği görülmüştür. Sonuç olarak, tez kapsamında geliştirilen lignin ve NKS içeren tek ve iki katmanlı PLA bazlı kompozit filmler; çevre dostu, aktif ve biyobazlı ambalaj filmlerinin geliştirilmesi açısından önemli bir potansiyel sunmaktadır.

Polylactic acid (PLA) is a promising polyester for food-packaging applications owing to its ease of processing, low production cost, biocompatibility, and biodegradability. However, despite these advantages, PLA exhibits several limitations, including brittleness, low thermal resistance, slow crystallization rate, low elongation at break and tensile strenght, and insufficient water vapor and oxygen barrier properties. In this thesis, in order to overcome these drawbacks, mono-layer and multilayer bio-based active nanocomposite films were developed and characterized by incorporating cellulose nanocrystals (NCC) and lignin (L) into the PLA matrix. Accordingly, monolayer films, including control PLA, PLA–NCC, PLA–L, and PLA–NCC–L, along with bilayer films including PLA–NCC/L and PLA–NCC/L–NCC, were fabricated by the solution casting method. For the preparation of monolayer films, PLA was dissolved in chloroform with 5 wt% polyethylene glycol (PEG 400) as a plasticizer, and successive magnetic stirring and ultrasonication steps were applied to obtain homogeneous film structures. PLA–NCC films contained 2 wt% NCC, PLA–L films contained 2 wt% lignin, and PLA–NCC–L films were prepared with the combined addition of 2 wt% NCC and 2 wt% lignin. The prepared film-forming solutions were cast into petri dishes and dried for 24 hours at 25 ± 2 °C and 50 ± 5% relative humidity to obtain the films. For the preparation of bilayer films, the first layer was based on PLA–NCC, whereas the second layers were obtained from lignin and lignin–NCC dispersions prepared in 0.01 M sodium hydroxide solution, followed by prolonged stirring and ultrasonication. During bilayer film fabrication, the first layer was subjected to a pre-drying step for 18 hours at 25 ± 2 °C and 50 ± 5% relative humidity. After casting the second layer, the films were further dried under the same temperature and relative humidity conditions for 24 hours. The developed mono- and bilayer composite films were comprehensively evaluated in terms of physical (thickness, color, solubility, swelling index, and water contact angle), structural and morphological (FTIR, XRD, and FE-SEM), thermal (DSC), mechanical (tensile strength, elongation at break, and heat-seal strength), barrier (water vapor permeability, light transmittance, and opacity), and active (antioxidant and antimicrobial) properties. Film thickness ranged from 31 to 56 µm, and the incorporation of NCC and/or lignin significantly increased thickness (p<0.05). Regarding optical performance, presence of lignin and the bilayer design were the dominant factors: while the control PLA exhibited the highest lightness (L* = 94.54), the bilayer PLA–NCC/L–NCC film showed the lowest lightness (L* = 55.28) and the greatest overall color difference (ΔE = 54.66). This behavior was attributed to enhanced light absorption in the visible region by chromophoric groups in lignin. In particular, the high concentration of lignin at the surface of bilayer films resulted in a pronounced brown coloration that was not observed in monolayer films. This surface-localized lignin distribution not only affected the optical properties but also enabled efficient absorption of UV radiation, thereby playing a critical role in enhancing the UV barrier performance of the films. No statistically significant differences in solubility and swelling index were observed among the monolayer PLA–NCC, PLA–L, and PLA–NCC–L films (p>0.05). In contrast, bilayered films, PLA–NCC/L and PLA–NCC/L–NCC films, exhibited significantly higher solubility and swelling indexes than the monolayer films (p<0.05). The highest solubility (6.72%) and swelling index (1.39%) values obtained for PLA–NCC/L–NCC. Consistent with these findings, the initial water contact angle of the control PLA was 68.5°, whereas only a slight increase was observed for the monolayer films; by contrast, the contact angle decreased on the lignin surface (56.3° at 0 min and 50.2° at 5 min) and the L–NCC surface (54.7° at 0 min and 49.9° at 5 min) in the bilayer films. FTIR analysis supported this observation by confirming the preservation of –OH stretching vibrations (3270-3330 cm⁻¹) and the presence of lignin-related aromatic/phenolic peaks in the L and L–NCC layers. These results indicate that polar functional groups, particularly hydroxyl groups, became more accessible at the L and L–NCC surfaces; the increased accessibility strengthened water–surface interactions and, consequently, lowered the contact angle. In monolayer films, FTIR spectra largely overlapped with the characteristic bands of control PLA. Nevertheless, the shift of the band at 1750 cm⁻¹ in the control PLA to 1748, 1746, and 1744 cm⁻¹ for PLA–NCC, PLA–L, and PLA–NCC–L films, respectively, indicates the formation of weak physical and chemical interactions between the fillers and the PLA matrix. In XRD analysis, the typical diffraction peaks of PLA were observed at 2θ = 14.9°, 16.9°, and 18.5°. The peak at approximately 22.4° in PLA–NCC confirmed the presence of NCC, whereas the reduced peak intensities in PLA–L and PLA–NCC–L suggested that the amorphous nature of lignin may partially suppress the crystalline phase. In the bilayer PLA–NCC/L–NCC film, the intensified cellulose-related peak at 22.8° indicated a more pronounced cellulose crystalline phase in this structure. FE-SEM observations showed that the control PLA had a smooth and homogeneous surface, while PLA–NCC exhibited a slight increase in surface roughness; PLA–L displayed microvoids; and PLA–NCC–L showed locally aggregated structures. In bilayer films, the L and L–NCC layers exhibited rougher, fibrous, and interpenetrated morphologies, whereas the PLA–NCC layer displayed a smoother and more continuous morphology. These findings confirm that the bilayer film structure was successfully formed and that the layers could be morphologically distinguished. Thermal analyses indicated that the glass transition temperature exhibited similar values (58.9–59.0 °C) across all film groups. While the melting temperature of monolayer films remained unchanged within the range of 155.6–155.8 °C, a slight increasing trend was observed in the melting temperatures of bilayer films. For control PLA, PLA–NCC, PLA–L, and PLA–NCC–L films, the cold crystallization temperature (Tcc) values were 108.5 °C, 106.1 °C, 113.5 °C and 111.1 °C, respectively. The ~2 °C decrease in Tcc upon NCC addition indicates a nucleating effect that facilitates PLA crystallization at lower temperatures. Conversely, lignin increased Tcc relative to the control PLA, which was attributed to partial restriction of chain mobility by the amorphous lignin structure, shifting crystallization to higher temperatures. Overall, the DSC results demonstrate that the additives did not alter the glass transition temperature of PLA, exerted a limited effect on the melting temperature, and predominantly influenced the cold crystallization behavior. In terms of mechanical properties, PLA–NCC films increased tensile strength from 33.03 MPa to 40.54 MPa while decreasing elongation at break to 1.73% comparing to control PLA. These results indicate that NCC effectively reinforced PLA but reduced ductility, leading to a more rigid structure. Although lignin did not provide a marked improvement in mechanical strength in monolayer structures, its combined use with NCC—particularly in bilayer designs—contributed positively to the balance between strength and flexibility. The bilayer PLA–NCC/L films exhibited the lowest tensile strength (28.86 MPa) and the highest elongation at break (2.69%), whereas the PLA–NCC/L–NCC films showed a relatively higher tensile strength (36.26 MPa) along with a relatively high elongation at break (2.32%). Additionally, all films were heat-sealable; successful sealing was achieved at 100 °C for monolayer films and at 95 °C for bilayer films. The lowest heat-seal strength was observed for PLA–NCC/L (165.98 N/m), whereas the highest value was obtained for the monolayer PLA–NCC–L film (315.44 N/m). Regarding barrier properties, the lowest water vapor transmission rate (WVTR) was obtained for the monolayer PLA–NCC film (5.08 g/m².24 hour), whereas the highest value was observed for bilayer PLA–NCC/L (7.40 g/m².24 hour). The control PLA film exhibited the lowest opacity (1.99 Abs/mm) and the highest light transmittance across the UV-C (77.2%), UV-B (82.1%), UV-A (85.5%), and visible (85.7%) regions. In contrast, lignin-containing monolayer films (PLA–L and PLA–NCC–L) showed significantly lower light transmittance in both the UV and visible regions compared to control PLA and PLA–NCC films, indicating that the UV barrier performance is primarily governed by the aromatic structure of lignin. The limited influence of nanocrystalline cellulose (NCC) on optical properties in monolayer lignin-containing films suggests that NCC plays a secondary role in light shielding. Bilayer films exhibited significantly higher opacity and markedly reduced UV transmittance compared to monolayer structures (p<0.05). Among all samples, PLA–NCC/L–NCC films displayed the highest opacity (16.39 Abs/mm) and the lowest light transmittance values, reaching complete UV-C blocking (0%) and substantial reductions in UV-B (0.2%), UV-A (1.0%), and visible light (15.6%). These findings demonstrate that the developed bilayer PLA-based films are particularly suitable for food-packaging applications where UV protection is prioritized over product visibility. Antioxidant activity results revealed that lignin-free PLA films exhibited negligible antioxidant activity. In contrast, lignin-containing monolayer films showed antioxidant activities in the range of 40–50%, while bilayer films reached values as high as 85%. In monolayer films, lignin was embedded within the polymer matrix, whereas in bilayer structures, the lignin-rich layer was in direct contact with the DPPH solution, allowing greater surface accessibility of active phenolic components, and thus enhancing antioxidant effectiveness. Antibacterial activity was not detected in any of the monolayer films. In contrast, bilayer PLA–NCC/L films exhibited the strongest antibacterial performance, with inhibition zones of 7.9 mm against Escherichia coli, 5.6 mm against Staphylococcus aureus, 2.1 mm against Salmonella typhimurium, and 2.5 mm against Bacillus subtilis (p<0.05). PLA–NCC/L–NCC films showed antibacterial activity against E. coli, S. aureus, and S. typhimurium, but no inhibition was observed for B. subtilis. Neither monolayer nor bilayer films exhibited antifungal activity against Candida albicans or Yarrowia lipolytica. However, both bilayer structures showed antifungal effects against Aspergillus niger, with inhibition zones of 5.6 mm for PLA–NCC/L and 4.6 mm for PLA–NCC/L–NCC films. Overall, the results demonstrate that NCC primarily governs mechanical performance in PLA-based composite films, whereas lignin plays a decisive role in optical properties as well as antioxidant and antimicrobial activities. The bilayer design enhanced surface accessibility of active components, enabling improved UV barrier performance and strengthened antioxidant and antimicrobial effects, and further demonstrated that surface properties can be tailored through a multilayer architecture. In conclusion, the mono- and bilayer PLA-based nanocomposite films containing NCC and lignin show potential for use as environmentally friendly, active, and biodegradable packaging films, as reflected by their improved mechanical properties, enhanced UV barrier performance, antioxidant and the antimicrobial properties observed in bilayer structures.
