Özet

Teobromin, kakao kaynaklı bir metilksantin olup adipogenez ve adipoz doku kahverengileşmesi üzerinde potansiyel etkiler göstermektedir. Bu çalışmanın amacı, teobrominin in vitro ve in vivo modellerde adipogenez ve adipoz doku kahverengileşmesi üzerindeki etkilerini değerlendirmektir. Çalışmanın in vitro aşamasında, 3T3-L1 preadipozit hücrelerine diferansiyasyon ve maturasyon süreçlerinde farklı dozlarda teobromin uygulanmış; adipogenez (PPARγ, C/EBPα, FABP4) ve kahverengileşme (UCP1, PGC-1α, PRDM16, CIDEA) ile ilişkili moleküler belirteçlerin gen ekspresyon ve protein düzeylerinin yanı sıra, hücre içi trigliserit birikimi ve hücre canlılığı düzeyleri analiz edilmiştir. İn vivo aşamada, altı haftalık C57BL/6J dişi fareler (n=24), adaptasyon sonrası koruma (n=12) ve iyileşme (n=12) gruplarına ayrılmış; her grup serbest teobromin ve kontrol alt gruplarına bölünmüştür. Koruma grubunda, obezite gelişimini önleyici etkiyi değerlendirmek amacıyla, yüksek yağlı diyetle eş zamanlı olarak 10 hafta boyunca haftada bir kez kuyruktan intravenöz yolla 20 µM serbest teobromin uygulanmıştır. İyileşme grubunda ise, 10 haftalık yüksek yağlı diyet ile obezite modeli oluşturulduktan sonra, teobrominin terapötik potansiyelini değerlendirmek amacıyla, izleyen 10 hafta boyunca aynı doz ve uygulama sıklığında serbest teobromin müdahalesi gerçekleştirilmiştir. Deney süresince farelerin vücut ağırlıkları haftalık olarak izlenmiştir. Çalışma sonunda serum, inguinal beyaz adipoz doku ve kahverengi adipoz doku örnekleri toplanmıştır. Serumda protein düzeyleri (UCP1, PGC-1α, PPARγ) değerlendirilirken; adipoz doku örneklerinde adipogenez ve kahverengileşme ile ilişkili gen ekspresyon ve protein düzeyleri analiz edilmiştir. İn vitro bulgular, serbest teobrominin diferansiyasyon ve maturasyon süreçlerinde özellikle C/EBPβ ekspresyonu üzerinde doza ve zamana bağlı baskılayıcı etki gösterebildiğini, ancak adipogenez ve kahverengileşmenin geç dönem belirteçleri ile trigliserit birikimi üzerinde belirgin bir değişiklik oluşturmadığını ortaya koymuştur (p>0.05). İn vivo aşamada teobromin müdahalesine bağlı olarak farelerin vücut ağırlıklarında anlamlı bir fark gözlemlenmezken, koruma grubunda serbest teobromin uygulanan farelerin serumlarında UCP-1, PGC-1α ve PPARγ protein düzeylerinin kontrol grubuna göre arttığı (p<0.05); iyileşme grubunda ise serbest teobromin verilen farelerin kahverengi adipoz doku ve serumlarında PGC-1α protein konsantrasyonunun kontrol grubuna kıyasla daha yüksek olduğu saptanmıştır (p<0.05). Sonuç olarak, teobromin adipogenez ve adipoz doku kahverengileşmesinin düzenlenmesinde potansiyel bir fitokimyasal ajan olup, etki mekanizmalarının netleştirilmesi için ileride yapılacak olan çalışmalara ihtiyaç duyulmaktadır.

Theobromine is a cocoa-derived methylxanthine that exhibits potential effects on adipogenesis and adipose tissue browning. The aim of this study was to evaluate the effects of theobromine on adipogenesis and adipose tissue browning using in vitro and in vivo models. In the in vitro phase of the study, 3T3-L1 preadipocyte cells were treated with different doses of theobromine during the differentiation and maturation processes. Gene expression and protein levels of molecular markers related to adipogenesis (PPARγ, C/EBPα, FABP4) and browning (UCP1, PGC-1α, PRDM16, CIDEA), as well as intracellular triglyceride accumulation and cell viability levels, were analyzed. In the in vivo phase, six-week-old female C57BL/6J mice (n = 24) were divided into prevention (n = 12) and recovery (n = 12) groups following an adaptation period, and each group was further subdivided into free theobromine and control subgroups. In the prevention group, to evaluate the preventive effect on obesity development, 20 µM free theobromine was administered intravenously via the tail vein once weekly for 10 weeks concurrently with a high-fat diet. In the recovery group, after the establishment of an obesity model with a 10-week high-fat diet, free theobromine was administered at the same dose and frequency for an additional 10 weeks to assess its therapeutic potential. Body weights of the mice were monitored weekly throughout the experimental period. At the end of the study, serum, inguinal white adipose tissue, and brown adipose tissue samples were collected. Protein levels (UCP1, PGC-1α, PPARγ) were evaluated in serum samples, while gene expression and protein levels related to adipogenesis and browning were analyzed in adipose tissue samples. The in vitro findings demonstrated that free theobromine exerted a dose- and time-dependent suppressive effect on C/EBPβ expression during the differentiation and maturation processes, but did not induce significant changes in late-stage markers of adipogenesis and browning or in triglyceride accumulation (p > 0.05). In the in vivo phase, no significant differences in body weight were observed following theobromine intervention; however, in the prevention group, serum levels of UCP-1, PGC-1α, and PPARγ proteins were higher in free theobromine–treated mice compared with the control group (p<0.05). In the recovery group, PGC-1α protein concentrations in brown adipose tissue and serum were also higher in free theobromine–treated mice than in the control group (p<0.05). In conclusion, theobromine is a potential phytochemical agent in the regulation of adipogenesis and adipose tissue browning; however, further studies are required to clarify its mechanisms of action.
