Özet

Amaç: Araştırmada yeşil çaya farklı oranlarda nar, kızılcık, karadut ve aronya ilave edilerek yeşil çayın fenolik madde ve antosiyanin açısından zenginleştirilmesi, antioksidan aktivitesinin artırılarak yeşil çayın fonksiyonel özelliğinin geliştirilmesi, biyoaktif bileşiklerinin in vitro gastrointestinal sistemde biyoerişilebilirliğinin belirlenmesi, yeşil çayın renk ve aroma bakımından tüketici beğenisinin artırılması amaçlanmıştır. Yöntem: : Araştırmada liyofilizasyonla kurutulduktan sonra öğütülen meyveler, öğütülmüş yeşil çaya %30, %40 ve %50 oranlarında ilave edilmiş ve ardından 80±1°C'de 5 dk infüze edilerek çay infüzyonları hazırlanmıştır. Meyveli yeşil çayların ve sade yeşil çayın (kontrol) toplam fenolik madde (TFM), toplam flavonoid (TF), toplam monomerik antosiyanin (TMA), titrasyon asitliği miktarları renk ve pH değerleri, antioksidan aktiviteleri (DPPH·, ABTS+·, FRAP, CUPRAC), fenolik profilleri ve duyusal özellikleri tespit edilmiştir. Ayrıca infüzyonlar in vitro gastrointestinal sistemde sindirime tabi tutularak mide ve bağırsak fazlarının TFM, TF, TMA miktarları, antioksidan aktiviteleri, fenolik profilleri ve bunların biyoerişilebilirlikleri yukarıda verilen yöntemlerle belirlenmiştir. Bunlara ilaveten yeşil çayın su, %80 etanol-su ve %80 metanol-su ekstraktlarının TFM ve antioksidan aktivileri tespit edilmiştir. Bulgular: Yeşil çay ekstraktlarının TFM miktarları (mg GAE/g) 84,82±0,90 (su ekstraktı)- 195,86±8,76 (etanol-su ekstraktı); TF miktarları (mg QE/g) 76,14±2,96 (su ekstraktı)- 178,28±1,22 (etanol-su ekstraktı); DPPH aktiviteleri (mM TE/100g) 74,77±2,10 etanol-su ekstraktı)– 87±0,70 (su ekstraktı); ABTS aktiviteleri (mM TE/100g) 114,80±6,55 (su ekstraktı)– 192,14±3,45 (metanol-su ekstraktı); CUPRAC aktiviteleri (mM TE/100g) 0,84±0,03 (su ekstraktı)– 1,50±0,048 (metanol-su ekstraktı); FRAP aktiviteleri (mM TE/100g) 5,09±0,164 (su ekstraktı) – 11,42±0,052 (etanol-su ekstraktı) olarak belirlenmiştir. Çay infüzyonlarının L*, a*, b* C* ve Hº değerleri sırasıyla: 58,53- 43,50; -1,93-15,02; 1,85-11,29; 2,81- 15,13 ve 7-99,7; pH değerleri 3,74-6,12, titrasyon asitliği miktarları %0,01-0,06 olarak tespit edilmiştir. Çay infüzyonlarının sindirim öncesi ve in vitro sindirim sonrası (mide ve bağırsak) sırasıyla TFM miktarları 660,22-1824,63; 274,59- 601,30; 227,64-340,25 mg GAE/L; TF içerikleri 426,91-1058,96; 216,19-501,01; 225,88-346,07 mg QE/L; antioksidan aktiviteleri DPPH ile 365,70-764,73; 128,19-270,14; 131,18-180,23 μM TE/ 100mL; ABTS ile 714,42-1533,95; 203,55-624,31; 222,32-323,85 μM TE/ 100mL; FRAP ile 427,58-853,82; 178,45-355,01; 47,74-75,31 μM TE/ 100mL; CUPRAC ile 693,87-1411,65; 303,99-586,21; 163,09-276,33 μM TE/ 100mL olarak belirlenmiştir. İnfüzyonların sindirim öncesi TMA miktarları 2,91-78,75 mg CY3-G/L arasında tespit edilirken, simüle sindirim sonrası mide ve bağırsak fazlarında antosiyanin saptanamamıştır. Çay infüzyonlarının sindirim sonrası mide ve bağırsak ortamlarındaki % biyoerişilebilirlikleri sırasıyla TFM 30,48-49,96; 18,66-36,06; TF 43,70-55,01; 32,91-52,94; DPPH 22,02-39,17; 17,16-48,86; ABTS 15,51-44,72; 20,91-31,76; FRAP 28,05-50,19; 8,25-11,58 ve CUPRAC 31,87-45,82; 19,40-24,03 olarak belirlenmiştir. Araştırmada analiz edilen gallik asit, klorojenik asit, kafeik asit, epigallokateşin, kateşin, epikateşin ve rutin çay infüzyonlarında tespit edilirken, gallik asit mide ve bağırsak fazlarında tespit edilememiş, diğer fenolik bileşikler de farklı fazlarda farklı miktarlarda belirlenmiştir. Sonuç: Bu çalışmada, fonksiyonel yeni ürünler olarak üretilen meyveli yeşil çayların yüksek miktarlarda TFM, TF ve TMA içerdikleri, fenolik bileşikler açısından zengin ve yüksek düzeyde antioksidan aktiviteye sahip oldukları belirlenmiştir. Meyve-yeşil çay infüzyonlarının, TFM ve TF içeriklerinin, antioksidan aktivitelerinin yeşil çaya göre daha düşük olduğu, ancak in vitro sindirim sonrasında mide ve bağırsak fazlarında bu bileşiklerin biyoerişilebilirliklerinin meyveli çay infüzyonlarında daha yüksek olduğu belirlenmiştir. Dolayısıyla bu bulgulardan, meyve ilavesinin yeşil çayın fonksiyonelliğini artırdığı söylenebilir. Bunun yanı sıra, piyasada yer alan bitki ve meyve çaylarına alternatif olabilecek özellikte, klasik yeşil çayların genel olarak sahip olduğu renk ve buruk tat gibi tüketici tarafından cazip olmayan özellikleri, meyve ilavesi sayesinde giderilerek daha canlı renk ve hoş aromaya sahip yeni bir fonksiyonel ürün geliştirilmiştir. Bu sayede, sağlık üzerine olumlu etkide bulunan fonksiyonel içecek yelpazesine yeni bir alternatif sunulmuştur.

Purpose: The study aimed to enrich green tea with phenolic compounds and anthocyanins by adding different proportions of pomegranate, cornelian cherry, mulberry, and aronia; to enhance its antioxidant activity and thereby improve its functional properties; to determine the bioaccessibility of its bioactive compounds in an in vitro gastrointestinal system; and to increase consumer acceptability of green tea in terms of color and aroma. Method: In the study, fruits were freeze-dried, ground, and added to powdered green tea at ratios of 30%, 40%, and 50%. The mixtures were then infused at 80 ± 1 °C for 5 minutes to prepare the tea infusions. The total phenolic content (TPC), total flavonoid content (TFC), total monomeric anthocyanin (TMA), titratable acidity, color and pH values, antioxidant activities (DPPH·, ABTS+·, FRAP, CUPRAC), phenolic profiles, and sensory properties of the fruit-enriched green teas and plain green tea (control) were determined. Furthermore, the infusions were subjected to in vitro gastrointestinal digestion, and the TPC, TFC, and TMA contents, antioxidant activities, phenolic profiles, and their bioaccessibility in the gastric and intestinal phases were evaluated using the methods described above. In addition, the TPC and antioxidant activities of green tea extracts prepared with water, 80% ethanol–water, and 80% methanol–water were also analyzed. Findings: The total phenolic content (TPC) of green tea extracts (mg GAE/g) was determined as 84.82 ± 0.90 (water extract) and 195.86 ± 8.76 (ethanol–water extract); total flavonoid content (TFC, mg QE/g) was 76.14 ± 2.96 (water extract) and 178.28 ± 1.22 (ethanol–water extract). The DPPH radical scavenging activities (mM TE/100 g) were found as 74.77 ± 2.10 (ethanol–water extract) and 87 ± 0.70 (water extract); ABTS activities (mM TE/100 g) ranged between 114.80 ± 6.55 (water extract) and 192.14 ± 3.45 (methanol–water extract); CUPRAC activities (mM TE/100 g) were 0.84 ± 0.03 (water extract) and 1.50 ± 0.048 (methanol–water extract); FRAP activities (mM TE/100 g) were determined as 5.09 ± 0.164 (water extract) and 11.42 ± 0.052 (ethanol–water extract). The L*, a*, b*, C* and Hº color values of tea infusions were measured as 58.53–43.50, −1.93–15.02, 1.85–11.29, 2.81–15.13, and 7–99.7, respectively. The pH values ranged from 3.74 to 6.12, and titratable acidity was found between 0.01–0.06%. Before and after in vitro digestion (gastric and intestinal phases), the TPC of tea infusions (mg GAE/L) was determined as 660.22–1824.63; 274.59–601.30; 227.64–340.25, respectively. TFC values (mg QE/L) were 426.91–1058.96; 216.19–501.01; 225.88–346.07, respectively. Antioxidant activities were measured as follows: DPPH 365.70–764.73; 128.19–270.14; 131.18–180.23 μM TE/100 mL; ABTS 714.42–1533.95; 203.55–624.31; 222.32–323.85 μM TE/100 mL; FRAP 427.58–853.82; 178.45–355.01; 47.74–75.31 μM TE/100 mL; CUPRAC 693.87–1411.65; 303.99–586.21; 163.09–276.33 μM TE/100 mL, respectively. The total monomeric anthocyanin (TMA) content of infusions before digestion was determined between 2.91–78.75 mg CY3-G/L, while no anthocyanins were detected after simulated gastric and intestinal digestion. The bioaccessibility percentages of tea infusions after gastric and intestinal digestion were as follows: TPC 30.48–49.96; 18.66–36.06; TFC 43.70–55.01; 32.91–52.94; DPPH 22.02–39.17; 17.16–48.86; ABTS 15.51–44.72; 20.91–31.76; FRAP 28.05–50.19; 8.25–11.58; CUPRAC 31.87–45.82; 19.40–24.03, respectively. Among the analyzed phenolic compounds, gallic acid, chlorogenic acid, caffeic acid, epigallocatechin, catechin, epicatechin, and rutin were identified in tea infusions. Gallic acid, however, was not detected in the gastric and intestinal phases, while the other phenolic compounds were found in different amounts across the digestion phases. Results: In this study, it was determined that the fruit-enriched green teas, produced as novel functional products, contained high levels of total phenolic content (TPC), total flavonoid content (TFC), and total monomeric anthocyanins (TMA), were rich in phenolic compounds, and exhibited strong antioxidant activity. Although the TPC, TFC, and antioxidant activities of the fruit–green tea infusions were lower compared to plain green tea, the bioaccessibility of these compounds in the gastric and intestinal phases after in vitro digestion was found to be higher in the fruit-enriched tea infusions. Therefore, these findings suggest that fruit addition enhances the functionality of green tea. Moreover, a new functional product was developed with the potential to serve as an alternative to commercially available herbal and fruit teas, overcoming the generally less appealing characteristics of conventional green teas—such as color and astringent taste—by providing a more vivid color and pleasant aroma through fruit enrichment. In this way, a new alternative has been introduced to the range of functional beverages that exert positive effects on health.
