öz AESCULUS HIPPOCASTANUM ÖZÜTLERİNDEN AESCİNİN MİKROZOMAL LİPİD PEROKSİDASYONU ÜZERİNDEKİ ETKİLERİ Sağdıçoğlu, Adviye Gülçin Yüksek Lisans, Biyokimya Bölümü Danışman: Doç. Dr. Nursen Çoruh Eylül 2000, 73 sayfa At kestanesi (Aesculus hippocastanum) ağaçları'ndan elde edilen özütlerin birçok ilaca hammadde olabildiği bilinmektedir. Saflaştırılmış ekstrelerden özellikle aescin, biyolojik sistemlerde serbest oksijen radikallerini tutarak, istenmeyen lipit peroksidasyon zincir reaksiyonun oluşumunu engellemektedir. Serbest radikal zincir reaksiyonu sonucu ortaya çıkan lipid peroksidasyonu, hücrenin yapısında ve fonksiyonunda istenmeyen değişikliklerin oluşmasına yol açmaktadır. Bu çalışmada, at kestanesi ekstresi aescini koyun karaciğer mikrozomlarında oluşan lipit peroksidasyonuna etkisi Thiobarbituric acid testi ile incelenmiştir. Protein konsantrasyonu, tampon çözelti türü ve pH değeri ve reaksiyonun zamanı açısından optimize edilen sistemde 0.9 mg/ml mikrozomal protein konsantrasyonu kullanılmıştır. 0.2 M Tris-HCI tamponçözeltisi pH 7.4 değerinde hazırlanmış ve reaksiyon karışımına 100 |j,M demir eklenerek lipit peroksidasyonu arttırılmıştır. 0.45mM aescin konsantrasyonunda ölçülen mikrosomal lipit peroksidasyonu 0.064 ±0.04 nmol MDA/dakika/mg protein değeri ile %50 inhibisyon göstermiştir.

ABSTRACT EFFECTS OF AESCIN FROM AESCULUS HIPPOCASTANUM EXTRACTS ON MICROSOMAL LIPID PEROXIDATION Sağdıçoğlu, Adviye Gülçin M. Sc, Department of Biochemistry Supervisor: Assoc. Prof. Nursen Çoruh September 2000, 73 pages Seeds of horse chestnut (Aesculus hippocastanum) are well known raw materials to get pure extracts for several drugs. These extracts from the chestnut seeds are known to play an important role in preventing the cancer cell proliferation and tumour growth. Especially, one of the purified extracts called aescin is a scavenger, in the biological systems, for oxygen free radicals that takes place in the unwanted chain reactions known as lipid peroxidation. Lipid peroxidation via free radical chain reactions leads to undesirable changes in the structure and function of cell constituents. In this study, inhibitory effect of aescin on lipid peroxidation was examined by Thiobarbituric acid test on sheep liver microsomal membranes. In optimised experimental conditions of 0.9 mg/ml protein, 0.2 M Tris-HCI buffer at pH 7.4 and 30 minutes induction by 100^M iron, antioxidative iiieffects of aescine on microsomal lipid peroxidation was measured as 0.064 ± 0.04 nmol MDA/min/mg of protein with 0.45 mM aescin which is corresponding to 50% inhibition.
