Özet
Yaygın kullanıma sahip lezzet verici bir gıda katkı maddesi olan monosodyum glutamat (MSG) gıda otoriteleri tarafından güvenli kabul edilmekle birlikte patofizyolojik ve toksikolojik etkileri ile bağırsak mikrobiyotasını değiştirebilmektedir. Bu nedenle, monosodyum glutamatın yavru sıçan bağırsak mikrobiyotası üzerindeki etkilerinin ve Lactobacillus delbrueckii subsp. bulgaricus'un bu etkiler üzerindeki düzenleyici potansiyelinin incelenmesi amaçlanmıştır. Çalışmada altı haftalık sekiz dişi ve sekiz erkek sıçandan oluşan dört grup bulunmaktadır. İlk grup kontrol grubu olup, ikinci gruba MSG (30 mg/kg/gün), üçüncü gruba Lactobacillus delbrueckii subsp. bulgaricus (LB/107 kob/gün), dördüncü gruba MSG (30 mg/kg/gün) ve LB (107 kob/gün) ile müdahale edilmiştir. Müdahaleler içme suyuna ilave edilerek yapılmıştır. Uygun bakım koşullarında on iki haftalık müdahale boyunca sıçanların haftalık olarak vücut ağırlığı ve su tüketimi verileri ile dışkı örnekleri toplanmıştır. Dışkı örneklerinden DNA izolasyonu yapıldıktan sonra yeni nesil dizileme yöntemi yardımıyla taksonomik sınıfların tespiti sonucu dışkı mikrobiyota kompozisyonu belirlenmiştir. LB grubunda yer alan erkek sıçanların vücut ağırlığı kazanımı kontrol grubuna kıyasla anlamlı şekilde azalma eğilimi göstermiştir (p<0,001). Deney sonunda dışkı mikrobiyotası Firmicutes bolluğu kontrol grubu erkek sıçanlarda artış gösterirken dişi sıçanlarda azalmıştır. MSG grubunda Firmicutes bolluğunda artış görülürken, LB grubu dişi sıçanlarda belirgin değişim olmamış, erkek sıçanlarda ise azalma görülmüştür. MSG+LB grubu sıçanlarda Firmicutes bolluğu deney sonunda azalmıştır. Bacteroidetes bolluğu deney sonunda kontrol ve MSG grubunda azalmış, LB grubunda ise artmıştır. MSG+LB grubu erkek sıçanlarda Bacteroidetes bolluğunda bir artış görülürken, dişi sıçanlarda bir miktar azalma gözlenmiştir. Actinobacteria bolluğu kontrol, MSG ve MSG+LB gruplarında azalmış, LB grubu erkek sıçanlarda belirgin değişim göstermezken dişi sıçanlarda bir miktar artmıştır. Dışkı mikrobiyotası Proteobacteria bolluğu kontrol grubu erkek sıçanlarda belirgin değişim göstermezken, dişilerde artmıştır. MSG ve MSG+LB grubunda deney sonunda Proteobacteria bolluğu artmış, LB grubu erkek sıçanlarda artış görülürken dişi sıçanlarda belirgin bir değişiklik görülmemiştir. Dışkı mikrobiyotası Firmicutes/Bacteroidetes oranı kontrol ve MSG gruplarında artış gösterirken, LB ve MSG+LB gruplarında azalmıştır. Bakteri çeşitliliğinin bir göstergesi olan Shannon çeşitlilik indeksi değerleri MSG (p<0,05) ve MSG+LB grubunda yer alan sıçanlarda azalmış, LB grubundaki sıçanlarda ise artmıştır (p<0,05). Bu sonuçlar, MSG ve Lactobacillus delbrueckii subsp. bulgaricus'un bağırsak mikrobiyotası üzerinde etkilerinin olduğunu göstermektedir. MSG ve Lactobacillus delbrueckii subsp. bulgaricus'un bağırsak sağlığı üzerindeki etkilerinin bilinmesi ve tüm paydaşlarla işbirliği içinde gerekli stratejilerin uygulanması halk sağlığının korunması açısından önemlidir.

Monosodium glutamate (MSG), a widely used flavouring food additive, is considered safe by food authorities. However, it can alter the intestinal microbiota with its pathophysiological and toxicological effects. MSG in processed foods can affect the intestinal microbiota with pathophysiological and toxicological effects. Therefore, this study aimed to investigate the effects of monosodium glutamate on the intestinal microbiota of rat pups and the regulatory potential of L. bulgaricus on these effects. The study had four groups of eight female and eight male rats aged six weeks. The first group was the control group; the second group received MSG (30 mg/kg/day), the third group received L. bulgaricus (LB/107 cfu/day), and the fourth group received MSG (30 mg/kg/day) and LB (107 cfu/day). MSG and LB were added to drinking water. Body weight, water consumption data, and faecal samples were collected weekly throughout the twelve-week intervention period under appropriate care conditions. After DNA isolation from faecal samples, the faecal microbiota composition of taxonomic units was determined using the next-generation sequencing method. The weight gain of male rats in the LB group tended to decrease significantly compared to the control group (p<0.001). At the end of the experiment, the abundance of Firmicutes in the faecal microbiota of rats increased in male rats of the control group and decreased in female rats. While there was an increase in Firmicutes abundance in the MSG group, there was no significant change in the LB group female rats, and a decrease was observed in male rats. Firmicutes abundance in MSG+LB group rats decreased at the end of the experiment. Bacteroidetes abundance decreased in the control and MSG groups and increased in the LB group. An increase in Bacteroidetes abundance was observed in male rats of the MSG+LB group, while a slight decrease was observed in female rats. The abundance of Actinobacteria decreased in control, MSG and MSG+LB groups; did not change notably in the LB group male rats but increased slightly in the female rats. The abundance of faecal microbiota Proteobacteria did not present a notable change in male rats in the control group but increased in females. In MSG and MSG+LB groups, it was observed that the abundance of Proteobacteria increased at the end of the experiment. While an increase in Proteobacteria was observed in male rats of the LB group, no significant change was shown in female rats. In the MSG+LB group, Proteobacteria abundance increased at the end of the experiment. The faecal microbiota Firmicutes/Bacteroidetes ratio increased in the control and MSG groups but decreased in the LB and MSG+LB groups. Shannon diversity index values, which is an indicator of bacterial diversity, decreased in rats in MSG (p<0.05) and MSG+LB groups and increased in rats in LB group (p<0.05). These results suggest that MSG and Lactobacillus delbrueckii subsp. bulgaricus have effects on gut microbiota. It is important to know the effects of MSG and Lactobacillus delbrueckii subsp. bulgaricus on intestinal health and to implement the necessary strategies in cooperation with all stakeholders to protect public health.
