Özet
Amaç: Pankreas kanseri, kötü prognozu nedeniyle her iki cinsiyette de kanser ölümlerinin önde gelen yedinci nedenidir. İlaç direnci nedeniyle; yüksek metastaz ve tümör nüksü, pankreas kanseri tedavisinde zorluklara yol açmaktadır. Bu nedenle pankreas tümörü büyümesini engellemede, mevcut bulguların daha iyi anlaşılmasını sağlamak ve gelecekteki pankreas kanseri araştırmalarında yön bulmak amacıyla yeni hedefler arayan çalışmalar hızla ortaya çıkmaktadır. Tümör metastazı, hücre yapışmasını ve ECM'nin proteolitik bozunmasını içeren karmaşık bir süreçtir. MMP'ler, ECM proteinlerini parçalama ve tümör istilasını ve metastazı kolaylaştırmak için matriks molekülleri içindeki kriptik bölgeleri açığa çıkarma yetenekleriyle karakterize edilir. CTHRC1'in yüksek ekspresyonunun, tümör yayılımı, istilası, göçü, yapışması ve metastazının pozitif düzenlenmesi yoluyla tümör oluşumunu ve gelişimini teşvik ettiği düşünülmektedir. Bu sebeple; CTHRC1, pankreas kanserinin komşu organlara yayılmasını engellemede umut verici bir hedef olarak görünmektedir. Bu çalışmada; PANC-1 ve BXPC-3 insan pankreas kanseri hücrelerinde, CRISPR/Cas9 genom düzenleme teknolojisi ile CTHRC1 genini susturup gen ifadesini regüle ederek; MMP ifadeleri ve tümörün metastatik kapasitesindeki değişimi karşılaştırmalı olarak incelemek amaçlanmıştır. Yöntem: Çalışmanın ilk basamağında PANC-1 ve BXPC-3 hücre hatlarında; CRISPR/Cas9 teknolojisini en doğru ve kolay şekilde kullanabilmek amacıyla in vitro transkripsiyon ve tarama çalışmaları gerçekleştirilerek CTHRC1 geni üzerinde hedef bölgede etkin sgRNA dizisi belirlenmiş ardından genom düzenleme protokolleri uygulanmıştır. CTHRC1 geni üzerinde CRISPR/Cas9 teknolojisi ile meydana getirilen düzenleme dizi analizi ve erime eğrisi analizi yöntemleri ile doğrulanmıştır. Çalışmanın ikinci basamağında hedef bölgede meydana getirilen düzenlemenin pankreas kanserinde MMP ifadeleri ve tümörün metastatik kapasitesinde meydana getirdiği değişim; gen ifade analizleri, hücre invazyon ve proliferasyon çalışmaları ile analiz edilmiştir. Bulgular: BXPC-3 hücre hattında hedef bölgede çift zincir kırığına bağlı çerçeve kayması mutasyona sebep olan indellerin varlığı ortaya konmuştur. Buna bağlı olarak; BXPC-3 hücrelerinin invazyon oranının azaldığı (p<0.05), CTHRC1 geni ve MMP gen ifadelerinin azaldığı (p<0.05), hücrelerin proliferasyon yeteneklerinin inhibe edildiği gözlenmiştir. Sonuç: CTHRC1'in pankreas kanserinde hücre göçü, invazyonu ve proliferasyonunda rol aldığı gösterilmiştir. Metastazdaki rolü nedeniyle CRISPR/Cas9 teknolojisi ile CTHRC1'in hedeflenmesinin pankreas kanseri tedavisinde potansiyel bir strateji olabileceği görüşündeyiz.

Objective: Pancreatic cancer is the seventh leading cause of cancer death in both genders due to its poor prognosis. Due to drug resistance; High metastases and tumor recurrence pose difficulties in the treatment of pancreatic cancer. Therefore, studies looking for new targets in inhibiting pancreatic tumor growth are emerging rapidly to provide a better understanding of current findings and to guide future pancreatic cancer research. Tumor metastasis is a complex process involving cell adhesion and proteolytic degradation of the ECM. MMPs are characterized by their ability to cleave ECM proteins and expose cryptic sites within matrix molecules to facilitate tumor invasion and metastasis. High expression of CTHRC1 is thought to promote tumorigenesis and development through positive regulation of tumor spread, invasion, migration, adhesion and metastasis. Therefore; CTHRC1 appears to be a promising target in inhibiting the spread of pancreatic cancer to neighboring organs. In this study; By silencing the CTHRC1 gene and regulating gene expression in PANC-1 and BXPC-3 human pancreatic cancer cells, with CRISPR/Cas9 genome editing technology; It was aimed to comparatively examine the change in MMP expressions and metastatic capacity of the tumor. Method: In the first step of the study, in PANC-1 and BXPC-3 cell lines; In order to use CRISPR/Cas9 technology in the most accurate and easiest way, in vitro transcription and screening studies were performed and the effective sgRNA sequence was determined in the target region on the CTHRC1 gene, and then genome editing protocols were applied. Editing of the CTHRC1 gene with CRISPR/Cas9 technology was confirmed by sequence analysis and melting curve analysis methods. In the second step of the study, the change in the MMP expressions and metastatic capacity of the tumor in pancreatic cancer by the regulation in the target region were analyzed by gene expression assays, cell invasion and proliferation studies. Results: The presence of indels that cause frameshift mutations due to double chain break in the target region in the BXPC-3 cell line were revealed. Consequently; It was observed that the invasion rate of BXPC-3 cells decreased (p<0.05), the expression of CTHRC1 gene and MMP gene decreased (p<0.05), and the proliferation abilities of the cells were inhibited. Conclusion: CTHRC1 has been shown to be involved in cell migration, invasion and proliferation in pancreatic cancer. We believe that targeting CTHRC1 with CRISPR/Cas9 technology may be a potential strategy in the treatment of pancreatic cancer due to its role in metastasis.
