Cytochrome P450 enzymes have been vital as a worldwide basic enzyme family, which are found in several types of living organisms. They are involved in numerous activities on living ones such as, metabolic pathways and lots of biosynthesis. Whilst they are dealing with their roles, as expected, they are working with some other proteins and enzyme systems to carry out all of these roles. One of these complementary proteins is, called as, NADPH Cytochrome Oxidoreductase (CYPOR) which transports electrons from NADPH to CYP enzymes to help their activities by electron donation. Although this process is not solely consisting of CYPOR and CYP enzymes, CYPOR has critical importance on these activities. It has been thought that, for this reason, knowing of both CYPs and CYPORs features, mechanism of actions and characteristics may lead to better understanding of some disease that are linked to these enzymes. Even more, inhibition of these enzymes could also provide better treatment strategies against various problems including, several cancer types and fungal-parasitic diseases. Our aim is to detect some inhibitors for CYP 1 enzymes and reductase enzyme and bring them into light for future studies. For this reason, we tried to test some compounds on human CYP 1A1 / 1B1 enzymes and yeast NADPH Cytochrome Oxidoreductase Enzymes ( which were previously expressed in yeast expression systems) to find out whether they have any inhibition effect on these enzymes. Pongamia pinnata, Glycyrrhiza glabra and broccoli plant extracts were tested on Fluorometric CYP inhibition assay while several chemical compounds, tannic acid and quercetin were implemented on MTT Reductase absorbance inhibition assay. Our results showed that plant extracts, except broccoli, have indicated proper inhibition on both CYP enzymes. On the other hand, compounds 144, 149 and 151 has exposed considerable competitive inhibition and remarkable inhibition percentage on yeast microsomal reductase. Additionally, tannic acid has strong inhibition effect whilst 4-Phenylimidazole and diphenyliodonium chloride has slighter inhibition effect on yeast microsomal reductase.
