ABSTRACT Masters Thesis PRODUCTION OF INVERTASE ENZYME BY USING SACCHAROMYCES CEREVISIAE CELLS, IMMOBILIZATION OF INVERTASE CONTAINING CELLS, INVERT SUGAR PRODUCTION WITH IMMOBILIZED CELLS RamiAl-TAWEEL Ankara University Graduate School of Natural and Applied Sciences Department of Chemistry Supervisor : Assoc. Prof. Dr. Sibel SUNGUR Many enzymes obtained from microorganisms are characterized according to their structure and the mechanisms of the reactions they control. Invertase enzyme catalyses the hydrolyses of disaccharite, sucrose to basic monosaccharite glucose and fructose. In this study Saccharomyces cerevisiae ATTC 28786 strain has been utilised as a source of invertase enzyme. The optimum pH, temperature, incubation period and kinetic features of Saccharomyces cerevisiae cell invertase have been scrutinized and characterized. The gelatin immobilization of Saccharomyces cerevisiae cells have been activated according to chemical crosslinking and coupling methods. Chrome III acetate and chrome III sulphate have been utilised as crosslinkers. In this study different concentrations of chrome III acetate and chrome III sulphate were used to determine the optimum crosslinkers. The optimum concentrations for chrome III acetate and chrome III sulphate have been determined as 0.016 mol dm“3 and 0.008 mol dm”3, respectively. Within the scope of tins study analysing the features of immobilized Saccharomyces cerevisiae cell invertase the stability of temperature and pH, kinetic features plus reutilisation and storing stability have been sifted through. In the optimum pH of immobilized Saccharomyces cerevisiae cell invertase, a deviation from 4.6 to 7.2 has been determined according to cell enzyme.VI It has also been determined that the immobilized specimens obtained through the method developed in this study may be preserved in normal room temperature for 28 days and utilised at least 15 times while maintaining its features.
