Prof. Dr. Abdurrahman TANYOLAÇ In conventional ethanol fermentation, the rate of ethanol production and cell concentration decreases due to inhibitory effect of the product. Some alternative methods exist for removing ethanol from the broth during fermentation. One of the alternative methods is extractive fermentation that has been studied extensively. The use of an organic solvent phase in biocatalytic applications can provide both kinetic and thermodynamic advantages. When the product is continously removed from the solvent phase, biocatalyst activity will increase. However the solvent must have a high capacity for ethanol and must be biocompatibile. On the other hand, the distribution coefficients of the biocompatible solvents are relatively low. In this study, the solvents which have high distribution coefficients for ethanol were theoretically obtained by using experimental data and UNJJFAC method. As a solvent, n-decanol with distribution coefficient is 0.448 [(g/g)/(g/g)] was selected to use in extractive fermentation. In order to reduce the decanol toxicity S.cerevisiae cells wereIV immobilized in Ca-alginate. In addition to protect the cells from the water soluble decanol, sun flower oil in 10% to 20% (v/v) concenration range was added to immobilization media for furter protection. Moreover, to prevent the floating of the beads, A1203 was added to the media in different quantities (5%-20% w/v) Experiments were performed in 250 ml bioreactor with constant temperature and agitation rate. Toxic effect of the decanol (for VfeT. broth (ml)Arsoivent (ml) > 2/1) was complatly eliminated in the prence of 30% (v/v) sun flower oil and 5% (w/v) AI2O3.
