Özet
Bu çalışmada, Ankara, Erzurum ve Erzincan illerinden değişik yaş grubundan broiler piliç ve yumurta tavuklarına ait bağırsak içeriği, iç organlar ve kümes altlıklarından sırasıyla 50, 54 ve 50 adet olmak üzere toplam 154 örnek, Listeria türlerinin varlığı yönünden incelendi. izolasyon çalışmalarında ön zenginleştirme besiyeri olarak NB2 ve TPB; ikinci zenginleştirme besiyeri olarak LEB buyyonu ve izolasyon besiyeri olarak KA, MOX ile PALCAM; saf koloni elde etmek için de TA ve ACA besiyeri kullanıldı. İzole edilen suşların identifikasyonu amacıyla karbonhidrat fermentasyon testleri yapıldı ve hemolitik suşlann ayırımı için“çift CAMP testi”uygulandı. Fare, kobay, tavşan ve civcivlerde L monocytogenes suşlarının patojinetesi belirlendi. 50 adet bağırsak içeriği örneğinden 4 adet (%8) Listeria monocytogenes, 3 adet (%6) L welshimeri, 1 adet (%2) L murrayi olmak üzere 8 adet (%16) Listeria spp. izole edildi. 54 adet iç organdan, 7 adet (%12.96) L monocytogenes, 2 adet (%3.7) L murrayi, 2 adet (%3.7) L welshimeri, 2 adet (%3.7) L innocua ve 1 adet (%1.85) L grayi olmak üzere 14 adet (%25.92) Listeria spp. izole edildi. 50 adet kümes altlığından 5 adet (%10) L murrayi, 4 adet (%8) L innocua, 3 adet (%6) L monocytogenes ve 1 adet (%2) L grayi olmak üzere 13 adet (%26) Listeria spp. izole edildi. İncelenen 154 adet örnekten 14 adet (%9.09) Listeria monocytogenes, 8 adet (%5.19) L murrayi, 6 adet (%3.89) L innocua, 5 adet (%3.24) L welshimeri ve 2 adet (%.129) L grayi olmak üzere toplam 35 adet (%22.72) Listeria spp. izole ve identifıye edildi. Bu çalışmada izole ve identifıye edilen 14 adet L monocytogenes susunun 9'unun (%64.28) serotip-1, 5'inin (%35.71) ise serotip-4 olduğu belirlendi. L monocytogenes suşlarının disk diffüzyon yöntemi ile çeşitli antibiyotiklere karşı duyarlılıkları incelendi. Bu çalışmada elde edilen bulgulara göre tavuk eti ve ürünlerinin ülkemizde halk sağlığı yönünden potansiyel bir risk oluşturabileceği kanısına varıldı.

SUMMARY In this study, totally 154 samples that consist of 50, 54 and 50 samples of intestinal contents, internal organs and litter respectively were investigated for the presence of Listeria spp. in broilers and layers at different age that reared in Ankara, Erzurum and Erzincan provinces. In isolation studies NB2 and TPB were used as preenrichment broth and LEB was used as secondary enrichment broth and BA, MOX and PALCAM were used as isolation medium and also TA and ACA were used to obtained pure colony. To identificate the isolated strains carbonhydrate fermentation tests and to differentiate haemolytic strains“double CAMP test”were carried out. The pathogenicity of L monocytogenes strains was determined using mice, guineapigs, rabbits and chicks. Out of 50 intestinal contents, 4 (8%) L monocytogenes, 3 (6%) L welshimeri, 1 (2%) L munrayi, totally 8 (16%) Listeria spp., were isolated. Out of 54 internal organs, 7 (12.96%) L monocytogenes, 2 (3.7%) L murrayi, 2 (3.7%) L welshimeri, 2 (3.7%) L innocua and 1 (1.85%) L grayi, totally 14 (25.92%) Listeria spp., were isolated. Out of 50 litters, 5 (10%) L munrayi, 4 (8%) L innocua, 3 (6%) L monocytogenes and 1 (2%) L grayi, totally 13 (26%) Listeria spp., were isolated. Out of 154 tested samples, 14 (9.09%) L monocytogenes, 8 (5.19%) L murrayi, 6 (3.89%) L innocua, 5 (3.24%) L welshimeri and 2 (1.29%) L grayi, totally 35 (22.72%) Listeria spp., were isolated and identified. In this study, it was found that out of 14 L monocytogenes strains, 9 (64.28%) were belonged to serotype-1 and 5 (35.71%) were belonged to serotype-4. The antibiotic sensitivity of L monocytogenes strains against several antibiotics were tested using disc diffusion test. It was concluded that, in respect to results of this study, chicken meat and its products might be a potantial risk for public health in our country.
