Özet
Ill ÖZET Bu çaîişmada, Eczacılık Fakültesi, Kimya Mühendisliği bölümünde, sentetik olarak sentez edilen ve ilaç hammaddesi olarak kullanılması amaçlanan 2, 4, 5, tri-fenil imidazol ve dokuz türevinin mutajenik aktivitesi, Ames / Salmonella / Mikrozom test yöntemine göre araştırılmıştır. Laboratuvar çalışmalarında, sözü edilen test yöntemi, plate incorparation metodu ile uygulanmıştır. Test organizması olarak Salmonella typhimurium'un TA 98 ve TA 100 suşları kullanılmıştır. Deneyler sonucunda, histidin gereksinimi duyan oksotrof mutantlar, test bileşeni etkisiyle histidin yapabilme özelliğini kazanmıştır. TA 98 ve TA 100 suşlarının verdiği revertant koloni sayısına göre, TA 98 susu daha anlamlı sonuçlar vermiştir. Buna göre kimyasal test bileşenlerimizin, çerçeve kayması şeklinde direkt mutajenik etkili olduğu saptanmıştır. Aynı işlemler, metabolik aktivasyon enzimlerini içeren S9 bileşimi ile tekrarlandığında, daha önce tespit edilen revertant koloni sayılarında belirgin bir düşme olmuştur. Bu durumun da metabolik aktivasyon ile meydana geldiği kabul edilebilir.

IV SUMMARY In this ssudy (report), mutagenic activities of 2, 4, 5-tri- phenyl imidasole which is synthesised in the department of Chemical Engineering, in Facult, of Pharmacy artificially and is purpased for using as a raw material of drug (medicine), and its nine (9) derivatives has been investigated by using Ames / Salmonella / Microsome test. In laboratory, the test mentioned above has been accompanied with the metod of plate incorparation. The strains of Salmonella typhimurium TA 98 and TA 100 have been used as test organisms. As a conclusion of experiments, auxotroph mutants needed histidin have been succeeded in producing its own histidin by using the effect the test component. Comparing the number of revertant colonies of the strain TA 100, TA 98 had better results. According to these results, it was determined that our components of chemical tests have direct mutagenic effects in the form of frame shift. When the same process was applied to the compound of S9 which includes enzyms of metabolic activation, a significantly decrease in the number of revertant colonies determined before has been observed. And it can be considered that this case is the result of metabolic activation.
