Özet
Kolorektal kanser, dünya genelinde en sık görülen malignitelerden biri olup, inflamasyon ve oksidatif stres hastalığın gelişim sürecinde önemli rol oynamaktadır. Çörek otu yağı (Black Seed Oil, BSO), içerdiği timokinon ve diğer biyoaktif bileşenler nedeniyle antioksidan, antiinflamatuar ve potansiyel antikanserojen etkileriyle dikkat çekmektedir. Bu çalışmada, oral BSO uygulamasının azoksimetan/dekstran sodyum sülfat (AOM/DSS) ile oluşturulan deneysel kolorektal karsinogenez modeli üzerindeki olası koruyucu etkilerinin histopatolojik, oksidatif stres ve protein ekspresyon düzeyleri açısından değerlendirilmesi amaçlandı. Çalışmada C57BL/6 fareler negatif kontrol (NK: 200 mg/kg BSO (BioQuinone, Biocell İlaç, İstanbul, Türkiye)), pozitif kontrol (PK: AOM/DSS), düşük doz BSO (DD: AOM/DSS+ 30 mg/kg BSO) ve yüksek doz BSO (YD: AOM/DSS+ 200 mg/kg BSO) olmak üzere dört gruba ayrıldı. AOM/DSS protokolü ile deneysel kolorektal karsinogenez modeli PK, DD ve YD gruplarında oluşturuldu. BSO (%2 timokinon) uygulaması oral gavaj yoluyla 3 günde bir olacak şekilde gerçekleştirildi. Yapılan histopatolojik değerlendirmede gruplar arasında mukozal mimari, displastik değişiklikler ve toplam doku hasarı skoru açısından değerlendirildi. Ayrıca TAS, TOS ve OSI düzeyleri ölçüldü; Bax, Bcl-xL, p53 ve phospho-Akt protein ekspresyonları Western blot yöntemiyle analiz edildi. Histopatolojik değerlendirmelerde; BSO uygulanan DD ve YD gruplarında, PK grubuna kıyasla mukozal mimarinin daha iyi korunduğu ve histopatolojik hasarın daha sınırlı olduğu görüldü. Bu iyileşmenin her iki tedavi grubunda izlenmekle birlikte, doz artışıyla daha belirgin hâle geldiği saptandı. Toplam histopatolojik hasar skor değerlendirmesinde, BSO uygulanan DD ve YD gruplarında histopatolojik skorların PK grubuna göre anlamlı xiii düzeyde daha düşük olduğu (p<0,001) görülmüştür. TAS değerleri bakımından gruplar arasında anlamlı fark saptanmazken, TOS ve OSI değerlerinde anlamlı farklılık bulundu. BSO uygulanmayan PK grubunda TOS ve OSI değerlerinin diğer gruplara göre anlamlı düzeyde daha yüksek olduğu saptandı (sırasıyla p=0,009 ve p=0,002). Yapılan protein ekspresyon analizlerinde Bax ekspresyonu açısından gruplar arasında anlamlı fark saptanmazken; p53, Bcl-xL ve p-Akt ekspresyonlarında istatistiksel olarak anlamlı farklılıklar görüldü (sırasıyla p=0,016, p=0,016 ve p=0,024). Bcl-xL ve p53 ekspresyonlarının, BSO uygulanan DD ve YD gruplarında arttığı ve en yüksek değerlerin YD grubunda olduğu görüldü (sırasıyla p=0,016 ve p= 0,016). p-Akt ekspresyonundaki farklılığın ise özellikle YD grubundaki artışla ilişkili olduğu, diğer gruplar arasında ise anlamlı fark bulunmadığı saptandı. Bu çalışmadan elde edilen bulgular, oral BSO uygulamasının AOM/DSS ile oluşturulan deneysel kolorektal karsinogenez modelinde koruyucu etki gösterebileceğini düşündürmektedir. BSO uygulanan gruplarda, özellikle histopatolojik olarak mukozal mimarinin daha iyi korunması ve doku hasarının daha sınırlı olması, bu etkinin kolon mukozası üzerinde yapısal düzeyde destekleyici bir rol oynayabileceğini göstermektedir. TOS ve OSI değerlerinin BSO uygulanmayan PK grubunda diğer gruplara göre daha yüksek bulunması, AOM/DSS uygulamasının oksidatif yükü artırdığını; BSO uygulanan gruplarda ise bu artışın sınırlanmış olabileceğini düşündürmektedir. Protein ekspresyon analizlerinde Bax düzeylerinde anlamlı fark saptanmaması, BSO'nun etkisinin belirgin bir proapoptotik yanıt artışından çok hücresel sağkalım ve adaptif yanıt mekanizmaları üzerinden şekillenebileceğini düşündürmektedir. Özellikle YD grubunda p53, Bcl-xL ve p-Akt ekspresyonlarının artması, BSO'nun epitel hücre sağkalımı, hücresel stres yanıtı ve doku onarımı süreçleri üzerinde etkili olabileceğini desteklemektedir. Bu bulgular birlikte değerlendirildiğinde, BSO'nun AOM/DSS ile indüklenen kolorektal karsinogenez sürecinde oksidatif stresin azaltılması, mukozal mimarinin korunması ve hücresel sağkalım yanıtlarının düzenlenmesi gibi birbiriyle ilişkili mekanizmalar üzerinden kolon karsinogenezinden koruyucu etki gösterebileceği düşünülmektedir.

Colorectal cancer is one of the most common malignancies worldwide, inflammation and oxidative stress play important roles in the development of the disease. Black seed oil (BSO) has attracted attention due to its antioxidant, anti-inflammatory, and potential anticarcinogenic effects, owing to its thymoquinone and other bioactive components. This study aimed to evaluate the possible protective effects of oral BSO administration on an experimental azoxymethane/dextran sodium sulfate (AOM/DSS)-induced colorectal carcinogenesis model in terms of histopathological findings, oxidative stress, and protein expression levels. In this study, C57BL/6 mice were divided into four groups: negative control (NC: 200 mg/kg BSO (BioQuinone, Biocell İlaç, İstanbul, Türkiye)), positive control (PC: AOM/DSS), low-dose BSO (LD: AOM/DSS + 30 mg/kg BSO), and high-dose BSO (HD: AOM/DSS + 200 mg/kg BSO). The experimental colorectal carcinogenesis model was established using the AOM/DSS protocol in the PC, LD, and HD groups. BSO containing 2% thymoquinone was administered by oral gavage once every three days. In the histopathological evaluation, the groups were assessed in terms of mucosal architecture, dysplastic changes, and total tissue damage score. In addition, TAS, TOS, and OSI levels were measured; Bax, Bcl-xL, p53, and phospho-Akt protein expressions were analyzed by Western blot. Histopathological evaluations showed that mucosal architecture was better preserved and histopathological damage was more limited in the BSO-treated LD and HD groups compared with the PC group. Although this improvement was observed in both treatment groups, it became more pronounced with increasing dose. In the assessment of total histopathological damage scores, the LD and HD groups treated with BSO had significantly lower xv histopathological scores than the PC group (p<0.001). While no significant difference was found among the groups in terms of TAS values, significant differences were observed in TOS and OSI values. TOS and OSI values were significantly higher in the non-BSO-treated PC group than in the other groups (p=0.009 and p=0.002, respectively). Protein expression analyses showed no significant difference among the groups in Bax expression; however, statistically significant differences were observed in p53, Bcl-xL, and p-Akt expressions (p=0.016, p=0.016, and p=0.024, respectively). Bcl-xL and p53 expressions increased in the BSO-treated LD and HD groups, with the highest values observed in the HD group (p=0.016 and p=0.016, respectively). The difference in p-Akt expression was found to be particularly associated with the increase in the HD group, while no significant difference was detected among the other groups. The findings obtained from this study suggest that oral BSO administration may exert a protective effect in an AOM/DSS-induced experimental colorectal carcinogenesis model. In the BSO-treated groups, particularly the better preservation of mucosal architecture and the more limited tissue damage observed histopathologically indicate that this effect may play a supportive structural role in the colonic mucosa. The higher TOS and OSI values in the non-BSO-treated PC group compared with the other groups suggest that AOM/DSS administration increased oxidative burden, whereas this increase may have been limited in the BSO-treated groups. The absence of a significant difference in Bax levels in protein expression analyses suggests that the effect of BSO may be shaped through cellular survival and adaptive response mechanisms rather than through a marked increase in proapoptotic response. In particular, the increased expressions of p53, Bcl-xL, and p-Akt in the HD group support the possibility that BSO may affect epithelial cell survival, cellular stress response, and tissue repair processes. Taken together, these findings suggest that BSO may exert a protective effect against colorectal carcinogenesis during the AOM/DSS-induced process through interrelated mechanisms such as reduction of oxidative stress, preservation of mucosal architecture, and regulation of cellular survival responses.
