Özet

Bu çalışmada, neodimyum (Nd) tabanlı metal-organik çerçeve (MOF) yapıları kullanılarak elektrokimyasal bir biyosensör geliştirilmiş ve kanser hücrelerinin seçici tespiti araştırılmıştır. Nd-MOF yapıları başarıyla sentezlenmiş ve yapısal ile yüzey özellikleri çeşitli karakterizasyon teknikleri ile doğrulanmıştır. Elde edilen yapılar folik asit (FA) ile fonksiyonelleştirilerek folat reseptörü aracılı hedefleme özelliği kazandırılmıştır. Geliştirilen biyosensörün elektrokimyasal davranışı döngüsel voltametri (CV), diferansiyel darbe voltametrisi (DPV) ve elektrokimyasal empedans spektroskopisi (EIS) teknikleri kullanılarak incelenmiştir. CV ve DPV analizlerinde, elektrot yüzeyinin MOF ile modifiye edilmesi ve hücre immobilizasyonu sonrasında pik akımlarında belirgin azalmalar gözlenmiş, bu durum elektrot yüzeyinde oluşan biyolojik tabakanın redoks probunun difüzyonunu sınırlandırması ve elektron transfer kinetiğini yavaşlatması ile ilişkilendirilmiştir. EIS analizleri, yüzey modifikasyon basamakları ile birlikte yük transfer direncinin (Rct) arttığını göstermiştir. Özellikle HeLa hücrelerinin immobilize edildiği yüzeylerde daha yüksek Rct değerleri elde edilmesi, hücre bağlanmasının etkinliğini xix doğrulamıştır. Farklı sayılarda HeLa hücresi kullanılarak gerçekleştirilen ölçümlerde, hücre sayısı ile biyosensör yanıtı arasında geniş bir aralıkta güçlü bir doğrusal ilişki elde edilmiştir. Sensörün düşük hücre konsantrasyonlarında dahi algılama yapabildiği ve analitik performansının yüksek olduğu belirlenmiştir. Seçicilik analizlerinde, folat reseptörü pozitif HeLa hücrelerinin, folat reseptörü negatif A549 hücrelerine kıyasla daha belirgin elektrokimyasal sinyal değişimine neden olduğu gözlenmiştir. Bu durum, folik asit ile fonksiyonelleştirilmiş sensör yüzeyinin hedef hücrelere karşı yüksek seçicilik sergilediğini ortaya koymaktadır. Sonuç olarak, geliştirilen Nd-MOF/FA tabanlı elektrokimyasal biyosensörün kanser hücrelerinin seçici, duyarlı ve güvenilir tespitinde etkili bir platform sunduğu gösterilmiştir. Bu yaklaşımın, erken tanı uygulamaları ve hedefe yönelik biyosensör tasarımlarında kullanılabilecek potansiyele sahip olduğu değerlendirilmektedir.

In this study, an electrochemical biosensor was developed using neodymium (Nd)-based metal- organic framework (MOF) structures, and the selective detection of cancer cells was investigated. The Nd-MOF structures were successfully synthesised, and their structural and surface properties were verified using various characterisation techniques. The resulting structures were functionalised with folic acid (FA) to impart folate receptor-mediated targeting capability. The electrochemical behaviour of the developed biosensor was investigated using cyclic voltammetry (CV), differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. In CV and DPV analyses, significant decreases in peak currents were observed following the modification of the electrode surface with MOF and cell immobilisation. This was attributed to the biological layer formed on the electrode surface restricting the diffusion of the redox probe and slowing down the electron transfer kinetics. EIS analyses, meanwhile, demonstrated an increase in charge transfer resistance (Rct) alongside the surface modification steps. In particular, the higher Rct values obtained on surfaces where HeLa cells were immobilised confirmed the effectiveness of cell attachment In measurements carried out with varying numbers of HeLa cells, a strong linear relationship was observed across a wide range between cell count and biosensor response. It was determined that the sensor is capable of detection even at low cell concentrations and demonstrates high analytical performance. In selectivity analyses, it was observed that folate receptor-positive HeLa cells caused a more pronounced electrochemical signal change compared to folate receptor-negative A549 cells. This finding demonstrates that the folic acid-functionalised sensor surface exhibits high xxi selectivity towards target cells. In conclusion, it has been demonstrated that the developed Nd- MOF/FA-based electrochemical biosensor provides an effective platform for the selective, sensitive and reliable detection of cancer cells. This approach is considered to hold significant potential for early diagnosis applications and the development of targeted biosensor Technologies.
