Özet
Amaç: Farelerde pentilentetrazol (PTZ) ile oluşturulan epilepsi modelinde Mas reseptör (MasR) agonisti AVE 0991'in nöbet parametreleri, anksiyete-benzeri davranışlar, öğrenme-bellek işlevleri üzerine etkilerini incelemeyi ve ayrıca MasR, NR2B, BDNF mRNA ekspresyon değişiklikleri üzerine etkilerini araştırmayı amaçladık. Gereç ve Yöntem: Çalışmada, 71 adet erkek Swiss albino fare kullanıldı. PTZ (37 mg/kg, i.p.) ile kronik tutuşma (kindling) modeli oluşturuldu. Hayvanlar tutuştuktan sonra, saf kontrol, PTZ kontrol (salin), 3 mg/kg ve 10 mg/kg AVE 0991 olmak üzere dört grup oluşturuldu. Tutuşan farelere elektrokortikografi (ECoG) elektrotları takıldıktan 1 hafta sonra AVE 0991 veya salin tek doz (PTZ'den 30 dk önce) ve 14 gün boyunca intraperitoneal olarak uygulandı. Nöbetler, nöbet evresi, ilk miyoklonik jerk latansı, jeneralize nöbet latansı, jeneralize nöbet süresi, jeneralize nöbetteki spike sayısı ve toplam spike sayısı parametreleri açısından değerlendirildi. Uzun süreli AVE 0991 veya salin uygulamalarının ardından açık alan testi, yükseltilmiş artı labirent ve Y-labirent testleri uygulandı. Hayvanlara kardiyak perfüzyon yapıldıktan sonra beyin dokuları çıkarıldı. Korteks ve hipokampusta MasR, NR2B ve BDNF mRNA ekspresyonları qPCR ile analiz edildi. Veriler uygun istatistiksel testlerle değerlendirildi. p<0,05 anlamlı kabul edildi. Bulgular: Tek doz AVE 0991 uygulaması nöbet parametrelerinde anlamlı bir değişiklik oluşturmadı. Buna karşın, uzun süreli AVE 0991 uygulaması (10 mg/kg), PTZ kontrol (salin) grubuna kıyasla nöbet evresini, jeneralize nöbet süresini ve spike sayısını azaltırken; ilk miyoklonik jerk ve jeneralize nöbet latansını uzattı. Açık alan testinde, 10 mg/kg AVE 0991 uygulaması, PTZ'nin azalttığı merkezde geçirilen süre ile şahlanma sayısını ve süresini artırdı. Yükseltilmiş artı labirent testinde, 3 ve 10 mg/kg AVE 0991 uygulanması PTZ'nin azalttığı açık kollara giriş sayısı ve burada geçirilen süreyi artırdı. Y-labirent testinde, PTZ kontrol grubunda azalan spontan alternasyon yüzdesi, 10 mg/kg AVE 0991 ile yükseldi. Korteks ve hipokampusta, MasR ekspresyonu PTZ kontrol grubunda değişmezken, AVE 0991 (3 ve 10 mg/kg) uygulanan grupta arttı. PTZ kontrol grubunda, korteks ve hipokampusta, artan NR2B ve BDNF mRNA düzeyleri, 10 mg/kg AVE 0991 uygulanmasıyla anlamlı şekilde azaldı. Tartışma ve Sonuç: MasR agonisti AVE 0991, PTZ ile indüklenen tutuşma modelinde hem nöbetleri hem de eşlik eden davranışsal ve moleküler bozuklukları azalttı. Bu bulgular, MasR hedefli tedavilerin epilepside umut vadedebileceğini düşündürmektedir.

Objective: This study aimed to investigate the effects of the Mas receptor (MasR) agonist AVE 0991 on seizure parameters, anxiety-like behaviors, learning and memory functions, as well as its effects on MasR, NR2B, and BDNF mRNA expression changes in a pentylenetetrazol (PTZ)-induced epilepsy model in mice. Materials and Methods: A total of 71 male Swiss albino mice were used in the study. A chronic kindling model was established using PTZ (37 mg/kg, i.p.). After the animals were kindled, four groups were formed: naïve control, PTZ control (saline), AVE 0991 3 mg/kg, and AVE 0991 10 mg/kg. One week after electrocorticography (ECoG) electrodes were implanted in the kindled mice, AVE 0991 or saline was administered intraperitoneally either as a single dose (30 min before PTZ) or daily for 14 days. Seizures were evaluated in terms of seizure stage, first myoclonic jerk latency, generalized seizure latency, generalized seizure duration, spike count during generalized seizures, and total spike count. Following long-term AVE 0991 or saline administration, open field, elevated plus maze, and Y-maze tests were performed. After cardiac perfusion, brain tissues were collected. MasR, NR2B, and BDNF mRNA expressions in the cortex and hippocampus were analyzed using qPCR. Data were evaluated using appropriate statistical tests, and p<0.05 was considered statistically significant. Results: Single-dose administration of AVE 0991 did not produce a significant change in seizure parameters. In contrast, chronic AVE 0991 administration (10 mg/kg), compared with the PTZ control (saline) group, reduced seizure stage, generalized seizure duration, and spike count, while prolonging the latency to first myoclonic jerk and generalized seizure onset. In the open field test, administration of 10 mg/kg AVE 0991 increased the time spent in the center as well as the number and duration of rearing behaviors that had been reduced by PTZ. In the elevated plus maze test, administration of 3 and 10 mg/kg AVE 0991 increased both the number of entries into the open arms and the time spent there, which had been decreased by PTZ. In the Y-maze test, the percentage of spontaneous alternation, which was reduced in the PTZ control group, increased following treatment with 10 mg/kg AVE 0991. In the cortex and hippocampus, MasR expression remained unchanged in the PTZ control group but increased in the AVE 0991-treated groups (3 and 10 mg/kg). In the PTZ control group, elevated NR2B and BDNF mRNA levels in the cortex and hippocampus were significantly reduced following administration of 10 mg/kg AVE 0991. Conclusion: The MasR agonist AVE 0991 attenuated both seizures and the accompanying behavioral and molecular impairments in the PTZ-induced kindling model. These findings suggest that MasR-targeted therapies may hold promise for the treatment of epilepsy.
