ÖZET Yüksek Lisans Tezi ARABÎDOPSÎS THÂLÎANA BİTKİSİNDE SODYUM AZİD VE EDTA MUTAGENLERİ İLE MUTASYON ÇALIŞMASI Rodin MAZAHERİ Ankara Üniversitesi Fen Bilimleri Enstitüsü Zootekni Anabilim Dalı Danışman: Prof. Dr. Tahsin KESİCİ Bu çalışmada, sodyum azid (NaN3) ve etilen diamin tetra asetik asit (EDTA)' nın Arabidopsis i hali ana tohumlan üzerindeki mutagenik etkileri araştırılmıştır. Bu amaçla Arabidopsis thattana tohumlan bu mutagenlerin farklı dozları ile muamele edilmiş, çimlenme miktarı, ilk çiçeklerime zamanı, bitki boyu ve silika sayısı özellikleri dikkate alınmıştır. Bu özellikler bakımından mutagen uygulanan tohumlardan elde edilen ölçümler, kontrol gruplarından elde edilenlerle karşılaştı nlmışiır. Ele alınan dozlarda EDTA uygulanan ve uygulanmayan tohumlar arasında, incelenen Özellikler bakımından istatistiksel olarak önemli bir fark bulı NaN3' in çimlenme miktan ve silika sayısı üzerine mutagenik etkisi incelendiğinde, kontrol ve muamele grupları arasında istatistiksel olarak önemli farklar bulunmuştur. Bitki boyu bakımından ise, dozlar arasındaki boy farklılığı günlere göre değişmektedir. Yani doz x gün intcraksiyonu önemli bulunmuştur. ABSTRACT Master Thesis MUTATION STUDY ON ARABIDOPSIS THAUANA WITH SODIUM AZIDEANDEDTA Rodin MAZAHERİ Ankara University Graduate School of Natural and Applied sciences Department of Animal Science Supervisor: Prof. Dr. Tahsin KESİCİ The purpose of this study is to investigate the mutagenic effects of sodium azide (NaN3) and ethylene diamine tetra acetic asid (EDTA) on Arabidopsis thaliana seeds. The Arabidopsis thaliana seeds were treated with different dosages of these mutagens. To evaluate the mutagenic effects of these chemicals, germination amount, the first flowering time, plant height and silique number were taken into consideration. The measurements on these features obtained from the treated seeds were compared with those collected from the untreated seeds. For the dosages of EDTA used, there was no statistically significant difference in the features taken into consideration between the treated and untreated seeds. Regarding the mutagenic effect of sodium azide on the germination amount and silique number the results of analysis showed that there was a statistically significant differences between the treated and control groups. The plant height, differs according to the applied dosages with respect to days. In other words, dosages x day interaction was found to be significant. The dosages used were compared within the days of the experiment 2000, 42 pages
