Özet
Bu çalışma ile Türkiye'de doğal olarak yayılış gösteren Caryophyllaceae familyası Eremogone cinsine ait taksonların palinolojisi, tohum mikromorfolojisi ve moleküler filogenisi incelenmiştir. Eremogone örnekleri doğal yayılış alanlarından toplanmıştır. Tez çalışması kapsamında palinoloji analizleri kapsamında Eremogone taksonlarının ışık mikroskobu ile Polen Çapı, Por Çapı, İki Por Arası Uzaklık, Ekzin Kalınlığı ve Por Sayısı ölçümleri yapılmış, Taramalı Elektron Mikroskobu (SEM) ile polen ornamentasyonu tanımlanmış ve ornamentasyon üyelerine ait ölçümler gerçekleştirilip, mikrofotoğraflar alınmıştır. Tohum mikromorfolojisi analizleri ile olgun tohum tanelerinin şekli ve rengi incelenmiş, en, boy ve hilum boyutları ölçülerek kayıt edilmiştir. SEM ile tohum kabuğu ornamentasyonu tanımlanmış ve ornamentasyon üyelerine ait ölçümler gerçekleştirilip, mikrofotoğraflar alınmıştır. Elde edilen nicel veriler tablo haline getirilmiş, grafikler hazırlanmış, uygun İstatistiksel yöntemler ile taksonların birbirleri ile olan benzerlik ve farklılıkları bulunarak dendrogramlar oluşturulmuştur. Moleküler filogeni çalışmaları için nükleer ribozomal DNA (ITS) ve kloroplast DNA (rps16) gen bölgelerinin PCR amplifikasyonu ile dizi analizleri elde edilmeye çalışılmıştır. Sekans verileri ile taksonların cins içi filogenetik ilişkileri araştırılmıştır. Yapılan çalışmalar neticesinde Eremogone polenlerinin taksonomik farklılıkları belirlenmiş, Eremogone tohumlarının Tohum En, Tohum Boy, Hilum ve Tohum Sırt Şekli karakterleri ile taksonlar arası ayırt edilebileceği sonuçlarına varılmıştır. Moleküler filogeni çalışmaları kapsamında kloraplast DNA'nın rps16 gen bölgesinden elde edilen diziler ile oluşturulan Gamma (rate variation) GTR (Substitution Model) uygulayanan MrBayes programının ürettiği taksonomik ağaç Eremogone taksonlarının genel morfolojik özelliklerine paralel sonuçlar ortaya koymuştur. Caryophyllaceae familyasına ait çoğu cinste olduğu gibi Eremogone cinsi de stenopalinozdür ve polen tipi ayırt edici özellikler sunmamaktadır. Eremogone tohumları ise polenlere kıyasla özellikle boyutları ve Tohum Sırt Şekli ile cins içinde ayırt edici özellikler göstermektedir. Cinsin moleküler filogeni çalışmaları ile cinsin genel morfolojisine paralel filogenetik ağaçlar elde edilmiştir. Tez çalışması kapsamında, cinsin taksonomik problemlerinin çözümüne katkı sağlanmıştır.

This project examined the palynology, seed micromorphology and molecular phylogeny of the Caryophyllaceae family Eremogone genus naturally distributed in Turkey. Within the scope of the thesis study, palynology analyses and pollen diameter, pore diameter, distance between two pores, exine thickness and pore number measurements of Eremogone taxa were made via light microscopy. Pollen ornamentation was determined by Scanning Electron Microscope (SEM), measurements of ornamentation members were made and microphotographs were taken. With seed micromorphology analyses, the shape and color of mature seed grains were examined, and the width, length and hilum dimensions were measured and recorded. Quantitative data were tabulated, dendrograms were created by finding the importance of similarities and differences between species using appropriate statistical methods, and these results were recorded with graphs. For molecular phylogeny studies, sequence analyzes of nuclear ribosomal DNA (ITS) and chloroplast DNA (rps16) gene regions were attempted to be obtained by PCR amplification. With the sequence data, the phylogenetic relationships of the taxa within the genus were investigated and their taxonomic status was reviewed. As a result of the studies, taxonomic differences of Eremogone pollen were determined, and it was concluded that Eremogone seeds could be distinguished between taxa with differences in Seed width, Seed Length, Hilum and Seed Back Shape. Within the scope of molecular phylogeny studies, the taxonomic tree produced by the MrBayes program, which applies the Gamma (rate variation) GTR (Substitution Model) created with the sequences obtained from the rps16 gene region of chloraplast DNA, revealed results parallel to the general morphological characteristics of Eremogone taxa. Like most genera belonging to the Caryophyllaceae family, the Eremogone genus is stenopalinous and does not present any distinguishing features of pollen type. Some Eremogone seeds show distinctive features within the genus, especially with their size compared to pollen. Phylogenetic trees parallel to the general morphology of the genus were obtained. Within the scope of the project work, contributions were made to the solution of taxonomic problems of the genus.
