Özet
60 ÖZET Türkiye' deki Safkan At Irklarında Bazı Enzim Sistemlerinin Elektroforetik İncelenmesi Bu araştırmada; esteraz enzim sistemi yönünden, 105 Arap, 112 İngiliz ve alyuvar enzim sitemleri (Fosfoglukonat dehidrogenaz=PGD, Fosfogluko- mutaz=PGM, Fosfohkzo izomeraz=PHI) yönünden 125 Arap, 123 İngiliz atı incelenmiştir. Esteraz lokusundaki fenotipik variantların ortaya konulması için alkali poliakrilamit jel elektroforezi ile isoelektirik fokuslama (IEF) yöntemi birlikte kullanılmıştır. Alyuvar enzim sistemleri ise pH 7,2' de tek bir nişasta jel elektroforezi yöntemi ile tespit edilmiştir. Esteraz enzim sisteminde, FF, Fİ, FS, Gl, lı ve SS olarak altı fenotip Arap atlarında, FF, Fİ, II, SS ve FS olarak beş fenotip İngiliz atlarında tespit edilmiştir. Her iki at populasyonunda I genotipinin en yüksek genotip frekansına sahip olduğu anlaşılmıştır. Nitekim homozigot II fenotipinin görülme yüzdesi Arap atlarında % 80,73 ve İngiliz atlarında % 75,89 olarak saptanmıştır. Esteraz enzim sistemi yönünden her iki populasyonun da Hardy-Weinberg Dengesi'nde olduğu anlaşılmıştır. Alyuvar enzim sistemlerinden olan PGD enzim sitemine ait; FF, FS ve SS olarak üç fenotip tespit edilmiştir. FS fenotipinin fenotipik görülme yüzdesi İngiliz atlarında Arap atlarına göre oldukça yüksek bulunmuştur. PGM enzim sitemi için homozigot SS ve PHI enzim sistemi için homozigot II fenotipleri gözlenmiştir.61

62 SUMMARY The Electrophoretical Investigation of Some Enzyme System in Arabian and Throughbreed Horses in Türkiye In this study, esterase enzyme systems in 109 Arabian and 112 Throughbreed horses and erythrocyte enzyme systems (Phosphogluconate dehydrogenase=PGD, Phosphogluco mutase=PGM and Phoshekso isomerase=PHI) in 125 Arabian and 123 Thoroughbreed horses were investigated. Both alkaline polyacrylamide gel electrophoresis and isoelectric focusing (IEF) were used to identify phenotipic variants of esterase locus. Erythrocyte enzyme systems were analyzed with only a starch gel electrophoresis at pH 7.2. Six phenotypes as FF, Fl, Gl, II, FS, SS in Arabian horses and five phenotypes as FF, Fl, FS, II, SS in Throughbreed horses were determined in esterase systems. Genotype I had the highest frequency in both Arabian and Througbreed horses. The frequency of homozygot II phenotype was 80,73 % in Arabian horses and 75,89 % in Throughbreed horses. It was realized that both populations were in the Hardy-weinberg Balance according to the esterase enzyme systems. FF, FS and SS phenotypes were detected in Arabian and Thoroughbreed horses for PGD which is one of the erythrocyte enzyme systems. The percentage of FS in Throughbreed horses was quite higher than that Arabian horses. In all examined animals, homozygot SS for PGM and II for PHI were observed.63
