Özet
Polikistik over sendromu (PKOS), üreme çağındaki kadınlarda en sık rastlanan endokrin-metabolik hastalıklardandır. YAP proliferasyon ve organ büyümesi ile ilişkili bir proteindir ve hippo yolağının önemli bir transkripsiyonel ko-aktivatörü olarak bilinir. PKOSlu kadınlarda yapılan gen analizlerinde artmış YAP seviyeleri ve promoter geninin hipometilasyonu gösterilmiştir. Verteporfin bir benzoporfirin türevidir ve ayrıca bir YAP/TAZ yolağı inhibitörüdür. YAP/TEAD etkileşimini bozarak ve YAP'ın sitoplazmada sekestrasyonunu arttırarak YAP degredasyonunda önemli bir rol oynamaktadır. Bu çalışmada YAP inhibitörü verteporfin letrozolle indüklenmiş PKOS'lu sıçan modellerine 21. Gün boyunca uygulandı. Kontrol, PKOS ve Verteporfin grubu olmak üzere 3 ayrı grupta deney modeli oluşturulup deneyin 22. Gününde deney hayvanlarından overler ve kan örnekleri alınarak, overlerde histokimyasal ve immunohistokimyasal incelemeler; kan serumu örneklerinde ise FSH, LH, östradiol ve testosteron hormonlarının profillerinin incelenmesi yapılmıştır. Yapılan deney sonuçlarına göre verteporfin grubunda PKOS grubuna göre testosteron seviyesinde anlamlı bir düşme olmasına rağmen over histolojisinde bir iyileşme gözlenememiştir. Bu çalışmanın sonucu ileri zamandaki çalışmalar için ipucu olacaktır.

Polycystic ovary syndrome (PCOS) is one of the most common endocrine-metabolic diseases in women of reproductive age. YAP is a protein associated with proliferation and organ growth and is known as an important transcriptional co-activator of the hippo pathway. Gene analyzes of women with PCOS found increased levels of YAP and hypomethylation of the YAP promoter which contributes to the increased levels of YAP. Verteporfin is a benzoporphyrin derivative and is also an inhibitor of the YAP/TAZ pathway. It plays an important role in the degradation of YAP by disrupting the YAP/TEAD interaction and increasing the sequestration of YAP in the cytoplasm. In this study, the YAP inhibitor verteporfin was administered to rats with letrozole-induced PCOS for 21 days. Experimental model was created as 3 groups: Control, PCOS and Verteporfin, respectively. On the 22nd day of the experiment, blood samples and ovaries taken from study animals. Histochemical and immunohistochemical evaluations conducted for the ovaries. In the blood serum samples, the hormone profiles such as FSH, LH, estradiol and testosterone were analyzed. Although there was a significant decrease in testosterone levels in the verteporfin group compared to the PCOS group, no improvement in ovarian histology was observed. The outcome of this study will be a clue for future studies.
