Özet

Bu çalışmanın amacı, yaşları 32 ile 72 arasında değişen 7 erkek ve 8 kadından oluşan toplam 15 kolorektal kanserli hastadan alınan tümör ve normal doku örneklerinde, kanser patogenezinde etkili olan p53 tümör süpresör gen (17p13.1) delesyonu ile erbB2 (Her-2/neu) onkogeni (17q11.2-q12) amplifikasyonunu saptamak için ilgili lokuslara spesifik problar kullanılmıştır. Çalışmamızda aile öyküsü olmayan kolorektal kanserli 15 hastadan 2 tanesinde p53 geni delesyonu saptanmıştır. Delesyon saptanan her iki olgunun da histopatolojik tipi adenokarsinom ve cerrahi evresi de Dukes sınıflandırmasına göre Dukes B olarak belirlenmiştir. 15 kolorektal kanserli olgudan oluşan çalışma grubumuzda histopatolojik tipi adenokarsinom ve cerrahi evresi Dukes D olan sadece 1 olguda erbB2 (Her-2/neu) amplifikasyonu laboratuvar eşik değerleri sınırında bulunmuştur. Yaptığımız çalışmada p53 delesyonu ve erbB2 (Her-2(neu) amplifikasyonu saptanan olguların farklı olgular olduğu görüldü.

The aim of this study was evaluate the signal phenotypes of p53 tumor suppressor gene and erbB2 (Her-2/neu) oncogene that is effective in the pathogenesis of cancer in the normal anf tumor tissue samples of 15 patients of 7 male and 8 female between ages of 32 and 72 with colorcetal cancer. Therefore, to detect the deletion of p53 tumor suppressor gene (17p13.1) and amplificitaion of ile erbB2 (Her-2/neu) oncogene (17q11.2-q12), related locus spsesific probes were used. In our study, in two of 15 patients with colrectal cancer who dis not have familial history, p53 gene deletion was detected. The histopathological type of both cases was adenocarcinoma and the surgicak stages was Dukes B according to Dukes classification. I our study, group of 15 patients with coloractal carcinoma, in only one cases with the histopathological type of adenocarcinoma and surgical stage of Dukes D, erbB2 (Her-2/neu) amplification was detected on the limit of the laboratuary cur-off values. In the study, p53 deletion and erbB2 (Her-2/neu) amplification were detected on different cases.
