Bazı florokinolon antibiyotiklerle santral sinir sistemi ilaçlarının farmakolojik etkileşim özellikleri üzerine deneysel araştırmalar

Özet
IV l.OZ Farelerde gerçekleştirilen bu çalışmada siprofloksasinin motor aktivite testlerinin sonucuna göre teofilinin etkisini önemli derecede artırdığı (p<0.05), diğer üç antibiyotiğin ise belirgin bir etkileşim göstermediği, belirlenmiştir. Open field testine göre florokinolon grubu antibiyotiklerle teofilin arasında bir etkileşim belirlenememiştir. Yüzme testinin sonuçlarına göre ise, siprofloksasin ve ofloksasinin teofilinin etkisini önemli derecede antagonize ettiği (p<0.05) saptanmıştır. Etomidatın meydana getirdiği uyku süresini florokinolonlardan siprofloksasin ortalama olarak 150 saniye ofloksasin 118 saniye, pefloksasin 142 saniye ve norfloksasin 98 saniye uzatmıştır. Fakat bu artış sadece siprofloksasin için anlamlı bulunmuştur (P<0.05). Open field testinden elde edilen sonuçlar da siprofloksasinin, etomidatın genel hareket aktivitesini azaltıcı etkisini önemli derecede güçlendirdiğini göstermektedir. Bu iki sonuç birlikte değerlendirildiğinde, siprofloksasinin etomidatın etkilerini anlamlı bir şekilde artırdığı kanısına varılmaktadır. Ofloksasin ve pefloksasinin etomidatın uyku sürelerini artırıcı etkileri istatistik yönden önemsiz (P>0.05) bulunmasının yanında open field testleri de bu iki ilaçla etomidatın genel hareket aktivitesi üzerine belirgin bir etkileşme göstermediğini ortaya çıkarmaktadır. Norfloksasinin ise open field testine göre etomidatın etkilerini önemli derecede güçlendirdiği, fakat uyku süresi üzerinde önemli bir artış şekillendirmediği belirlenmiştir. Sonuç olarak, florokinolon grubu antibiyotiklerle (siprofloksasin, ofloksasin, pefloksasin ve norfloksasin) sağaltım gören veya profilaktik amaçla bu ilaçların uygulandığı hastalarda santral sinir sistemi ilaçlan (teofilin ve etomidat) kullanılırken oluşabilecek etkileşimlerin göz önünde bulundurulması ve azami dikkat gösterilmesi önerilmektedir.

2. ABSTRACT Experimental studies on the pharmacologic interaction of some fluoroquinolone antibiotics with central nervous system drugs In this study which was carried out on mice, it was observed that ciprofloxacin significantly increased the interaction of theophylline according to the motor activity test (p<0.05), whereas effects of other drugs were not seen in this test. According to open field test, there was no interaction between floroquinolone and theophylline. On the other hand the swimming test showed that ciprofloxacin and ofloxacin had significantly antagonized the interaction of theophylline (p<0.05). The sleeping time which followed administration of etomidate was prolonged 150, 118, 142 and 98 seconds by ciprofloxacin, ofloxacin, pefloxacin and norfloxacin respectively. But this increase was significant only for ciprofloxacin (p<0.05). This observation was supported by the data of open field test in which the general activity of animal was diminished by ciprofloxacin following etomidate application. Therefore, it can be concluded that ciprofloxacin increases the effects of etomidate. Ofloxacin and pefloxacin were not effective on sleeping time caused by etomidate. This data was also supported by open field test. According to open field test, norfloxacin increased the effect of etomidate but it had no effect on the sleeping time. In conclusion, it is adviced that one should be cautious while using these antibiotics of fluoroquinolone group (ciprofloxacin, ofloxacin, pefloxacin, norfloxacin) in treatment of patients or for prophylactic purpose when the patient is given central neural system drugs (teophylline, etomidate).
