Özet

ÖZET Taksol (paklitaksel) çeşitli insan tümörlerine karşı antitümöral aktiviteye sahip bir diterpendir. Taksol sitotoksisitesinin esas olarak mikrotubullerin stabilizasyonu sonucu tubulin polimerizasyonunun artması ile hücrelerin G2/M fazında birikmesi ile oluştuğu düşünülmektedir. Çünkü hücre siklusunun bu fazlarındaki hücreler radyasyona karşı çok duyarlı olduktan bilindiğinden taksolün direkt toksisitesine ilave olarak tümör hücrelerim radyasyona karşı duyarlılaşılrabileceği düşünülmüştür. Eğer bu doğru olsaydı, insan tümörlerinin tedavisi için taksol ve radyoterapinin birlikte uygulanması gerçeğini sağlayacaktı. Ancak doku kültüründe taksolün radyoduyarlılaştırıcı etkisini test eden çalışmalar eşit sonuçlar vermiştir. Bu hücre soylarının yaklaşık %50'si taksol ile radyasyona karşı duyarlılaşmıştır. Biz bu çelişkiyi açıklayabilmek amacıyla, taksol ile oluşan radyoduyarlılık için sistemik bir çalışmayı başlattık. Bu konuda, taksolün hücreleri radyoduyarlılaştırabilme yeteneğinin taksole bağlı olarak apoptosis oluşturma yeteneği ile ters orantılı olarak değişebileceği hipotezini ileri sürdük. Bu hipotezi test edebilmek için biz bu çalışmamızda yedi farklı insan tümör hücre soylarında taksole bağlı olarak oluşan apoptosis (morfolojik değerlendirme ile) ve klonojenik hücre sağkalımı ile radyoduyarlılık yeteneklerini değerlendirdik. Kullandığımız hücre soylarının yaklaşık yana radyoduyarlıhk gösterdi (astrositoma; G18, prostat karsinoma; PC3, DU-145), oysa diğer yansında böyle bir etki gözlenmedi (kolon karsinoma; HT29, servikal karsinoma; HeLa, prostat carcinoma; LNCAP). Genel olarak, taksol tedavisinden 48 saat sonra düşük apoptosis gösteren hüce soyları radyasyona karşı duyarlılaşırken, çok yüksek apoptotik indekse sahip olan hücrelerde böyle bir etkiye rastlanmadı. Bu sonuçlar taksol tedavisinden sonra apoptosis seviyesinin taksole bağlı olarak oluşan radyoduyarlılığı belirleyebileceğini düşündürmektedir. 60SUMMARY Taxol (paclitaxel) is a deterpene with antitumor activity against a variety of human neoplasms. Studies evaluating the radiosensitizing potential of taxol in cultured cells have been equivocal with only approximately 50% of the tested cell lines showing some degree of radiosensitization. To address this apparent discrepancy, we initiated a systemic study for taxol induced radiosensitization. We advanced the hypothesis that the ability of taxol to radiosensitize cells may be inversely correlated to efficiency with which the drug induces apoptosis. To test the above hypothesis we studied a number of human tumor cell lines for taxol induced apoptosis (morphological evaluation) and radiosensitization by evaluating colony forming ability. Approximately half of the cell lines showed radiosensitization (astrocytoma; G18, prostate carcinoma; PC3, DU-145, and lung carcinoma; A549), whereas the remaining showed no radiosensitization (colon carcinoma; HT29, cervical carcinoma; HeLa, prostate carcinoma; LNCAP). In general, cell lines with low apoptotic index 48h after taxol treatment showed a significant degree of radiosensitization, whereas the cell lines with high apoptotic index showed no radiosensitization. This results suggest that the level of apoptosis, after taxol treatment, may predict for taxol-induced radiosensitization. 61
