Özet

D vitamini, güneş ışığı ile deride sentezlenen, bir kısmı ise diyetle alınan, hormon benzeri bir vitamindir. Çalışmada, tavşanlara Lipopolisakkarit (LPS) uygulanarak oluşturulan endotoksemik sepsis modelinde, D vitamininin oksidasyon ürünleri, serbest radikal metabolizmasında rol alan antioksidan moleküller ve serbest radikal düzeyleri üzerine etkisinin araştırılması amaçlandı. Çalışmada materyal olarak 32 adet Yeni Zelanda cinsi tavşan kullanıldı. 15 günlük adaptasyon döneminden sonra tavşanlar iki gruba ayrıldı. Birinci grup D vitamini eklenmiş standart tavşan yemi ile ikinci grup ise D vitamini eklenmemiş tavşan yemi ile 24 hafta beslendi. 24. haftanın sonunda kan örnekleri alınarak plazmaları elde edildi ve D vitamini tavşana özel ticari ELISA kit ile ölçüldü. Yetersizlik tespit edildikten sonra her iki grup ikişer gruba daha bölünerek dört grup oluşturuldu. Grup I (Vit D-) D vitaminininden eksik tavşan yemi ile, Grup II (Vit D+) standart tavşan yemi ile 24 hafta beslendikten sonra vena auricularisden tek doz serum fizyolojik verildi. Grup III (Vit D- (+) LPS) D vitaminininden eksik tavşan yemi ile, Grup IV (Vit D+ (+) LPS) standart tavşan yemi ile 24 hafta beslendikten sonra vena auricularisden tek doz 150 μg/kg LPS verildi. Uygulama sonrasında plazma nitrik oksit (NO), malondialdehit (MDA), redükte glutatyon (GSH) düzeyleri kimyasal yöntemle, 25-hidroksivitamin D3 (25OHD), melatonin ve 8-hidroksi-2-deoksiguanozin (8-OHdG) düzeyleri ise ELISA yöntemi ile ölçüldü. Çalışmada GSH düzeyleri Grup II'e göre Grup III ve Grup IV 'de düşük (p<0,05), melatonin düzeyleri Grup I'e göre Grup II'de ve Grup III'e göre Grup IV'te yüksek (p<0,01), MDA düzeyleri Grup II'ye göre Grup I ve III'de yüksek (p<0,05), 8-OH düzeyleri Grup III'de Grup I ve Grup II'ye göre yüksek (p<0,01), NO düzeyleri ise Grup I'e göre Grup II ve Grup IV'de düşük, Grup II'ye göre Grup III'de yüksek (p<0,01) bulundu. Sonuç olarak, lipid peroksidasyon ürünü olan MDA ve NO'in D vitamininden eksik beslenen grupta yeterli D vitaminine sahip gruba göre istatistiksel olarak yüksek bulunması, önemli bir antioksidan molekül olan melatoninin ise D vitamininden eksik beslenen grupta yeterli D vitaminine sahip gruba göre istatistiksel olarak düşük bulunması D vitamininin antioksidan özelliğe sahip olduğunu desteklemektedir. Yeterli D vitaminine sahip gruplarda GSH düzeyleri hariç LPS verilmeden öncesi ile verildikten sonraki değerler arasında istatistiksel olarak bir fark bulunamaması D vitamininin endotoksemik sepsise karşı koruyucu olabileceğini göstermektedir. Ancak D vitamininden eksik yemle beslenen gruplarda da 8-OHdG düzeyleri hariç diğer parametreler arasında istatistiksel fark bulunamamıştır. Bu durum D vitaminin GSH düzeyini arttırarak değil de daha çok melatonin düzeyini arttırarak antioksidan sistemi desteklediği kanaatini uyandırmaktadır.

Vitamin D is a hormone-like vitamin synthesized at skin with sunlight while some of which is received via diet. The study intends researching impact of vitamin D on oxidation products, antioxidant molecules that take part in free radical metabolism and free radical levels in endotoxemic sepsis model created by applying Lipopolysaccharide (LPS) to rabbits. We used 32 New Zealand rabbits in the study as material. Following 15-day adaptation period, the rabbits were divided into two groups. For 24 weeks, first group was fed with standard rabbit feed to which vitamin D was added while second group was fed with rabbit feed to which vitamin D was not added. At the end of 24 weeks, blood samples were taken, plasmas were obtained and vitamin D was measured with rabbit-specific commercial ELISA kit. After deficiency was detected, both groups were divided into two further groups and four groups were created. After feeding for 24 weeks Group I with rabbit feed deficient in vitamin D (Vit D-), Group II with standard rabbit feed (Vit D+), we administered single dose physiological saline solution through vena auricularis. After feeding for 24 weeks Group III with rabbit feed deficient in vitamin D (Vit D- (+) LPS), Group IV with standard rabbit feed (Vit D+ (+) LPS) we administered single dose 150 μg/kg LPS through vena auricularis. Following the application, levels of plasma nitric oxyde (NO), malondialdehyde (MDA), reduced glutathione (GSH) were measured with chemical method, levels of 25-hydroxyvitamin D3 (25OHD), melatonin and 8- hydroxy -2- deoxyguanosine (8-OHdG) with ELISA method. During the study, levels of GSH were found low at Group III and Group IV in comparison to Group II (p<0,05), levels of melatonin high at Group II in comparison to Group I and at Group IV in comparison to Group III (p<0,01), levels of MDA high at Group I and III in comparison to Group II (p<0,05), levels of 8-OH high at Group III in comparison to Group I and Group II (p<0,01), levels of NO low at Group II and Group IV in comparison to Group I, high at Group III in comparison to Group II (p<0,01). In conclusion, presence of MDA and NO, lipid peroxidation products, statistically high at the group fed deficient from vitamin D in comparison to the group that has sufficient vitamin D, while presence of melatonin, an important antioxidant molecule, statistically low at the group fed deficient from vitamin D in comparison to the group that has sufficient vitamin D supports the fact that vitamin D has antioxidant properties. Lack of statistical difference among values before and after LPS administration, levels of GSH excluded, at groups that have sufficient vitamin D indicates that vitamin D may be protective against endotoxemic sepsis. However, we also did not find any statistical difference among other parameters, except for 8-OHdG levels at groups fed with feed deficient in vitamin D. This forms the opinion that vitamin D supports antioxidant system by increasing level of melatonin rather than level of GSH.
