Özet

Glioma yüksek ölüm oranına sahip kötü huylu beyin tümörlerinden biridir. Beyni dolaşım sisteminden ayıran kan beyin bariyeri gliomada kemoterapinin etkinliğini kısıtlamaktadır. Büyük moleküllü ilaçlar kan beyin bariyerini geçemezken, küçük moleküllü ilaçların sadece %2'si geçebilmektedir. Terapötiklerin yetersiz biyodağılımı glioma tedavisindeki en büyük sorunlardan biridir. Bu nedenle ilaç dağılımını arttırmak için çeşitli ilaç taşıma sistemleri geliştirilmekte hatta bu ilaçlar istenilen bölgeye hedeflenmek üzere fonksiyonelleştirilebilmektedirler. Yüzeylerine çeşitli ligantlar bağlanarak hedeflenen taşıma sistemleri özel olarak kan beyin bariyerinde ve glioma hücrelerinde bulunan reseptörlere bağlanabilirler. Böylelikle, ilacın kan beyin bariyerini aşması ve hastalıklı hücrelerde yoğunlaşması sağlanır. Bu çalışmanın amacı Angiopep-2 peptidi ile fonksiyonelleştirilerek, glioma kanser hücrelerinin yüzeyinde sağlıklı hücrelere kıyasla daha fazla bulunan LRP-1 reseptörlerine hedeflenmiş, Kafeik Asit Fenetil Ester (CAPE) yüklü PLGA nanopartiküllerinin sentezi, karakterizasyonu ve C6 Glioma hücreleri üzerindeki sitotoksik etkisinin incelenmesidir. Kafeik Asit Fenetil Ester (CAPE) molekülü yüklü PLGA nanopartiküller tekli emülsiyon çözücü uçurma yöntemiyle sentezlenmiştir. Daha sonra, üretilen nanopartiküllerin hedeflendirilmesi amacı ile, Angiopep-2 peptidi mikrodalga destekli katı faz peptid sentezi yöntemiyle sentezlenmiş ve CAPE yüklü PLGA nanopartiküllerin yüzeyine bağlanmıştır. Boyut ve yüzey yükü analizleri sonucunda nanopartiküllerin 176.4 nm boyutlarında, -10.8 mV zeta potansiyele sahip ve monodispers (PDI<0.3) olarak sentezlendiği görülmüştür. FT-IR spektroskopi, boyut ve yüzey yükü analizleriyle angiopep-2 peptidinin nanopartiküllerin yüzeyine konjugasyonunun başarıyla gerçekleştiği gösterilmiştir. Sentezlenen nanopartiküllerin biyolojik aktivite çalışmaları L929 fibroblast ve C6 glioma hatları üzerinde MTT yöntemiyle incelenmiştir. Sitotoksik aktivite çalışmaları sonucunda serbest CAPE, CAPE yüklü nanopartiküller ve peptid ile hedeflendirilmiş nanopartiküllerin IC50 değerleri sırasıyla 50, 45 ve 21 µM olarak bulunmuştur. L929 fibroblast hücre hattıyla yapılan çalışmalar sonucunda literatürle de uyumlu olarak serbest CAPE ve nanopartiküllerin biyouyumlu oldukları görülmüştür. Bu tez çalışması sonucunda elde edilen Angiopep-2 ile hedeflendirilmiş CAPE yüklü nanopartiküller, serbest CAPE molekülü ve CAPE yüklü PLGA nanopartiküllerine kıyasla C6 Glioma hücreleri üzerinde daha yüksek sitotoksik etki göstermiştir.

Glioma is one of the malignant brain tumors with a high mortality rate. Chemotherapy is one of the most important methods in glioma treatment, but the blood-brain barrier limits the effectiveness of chemotherapy. Drugs with big molecular weight cannot cross the blood brain barrier, only 2 percent of drugs with small molecular weight can pass. Inadequate biodistribution of therapeutics is the biggest problem in glioma treatment. Therefore, various drug delivery systems are being studied to increase the drug pharmacokinetics/dynamics. Also, using these systems, drugs can be functionalized to target to the desired diseased region in the body. Targeted drug delivery systems can specifically bind to the cell receptors on the blood-brain barrier and glioma cells. Thus, drugs can pass through the BBB and accumulate in the diseased cells and tissues. In this study polymeric drug delivery system was synthesized for targeting glioma cell line. For this purpose FDA approved PLGA polymer was used to synthesize spherical nanoparticles and targeted with angiopep-2 peptide. Angiopep-2 is a synthetic peptide and also a ligand for LRP-1 receptors in the BBB and glioma cells. LRP-1 receptors are overexpressed on the surface of the BBB and glioma cells compared to healthy brain and BBB cells. For this study, polymeric drug delivery systems were designed to target glioma cells and increase the bioavailability of the drug. Angiopep-2 peptide was synthesized by microwave assisted solid phase peptide synthesis method. After cleavage of peptide, angiopep-2 peptide was analyzed with LC-MS. Caffeic acid phenethyl ester is an anticancer molecule and active compound of the propolis. In this study CAPE molecule was encapsulated in PLGA nanoparticles by single emulsion solvent evaporation method. Then, Angiopep-2 peptide was conjugated to the surface of CAPE loaded PLGA nanoparticles via EDC crosslinking agent. Characterization of the targeted nanoparticles were done with dynamic light scattering, FT-IR spectroscopy and SEM analysis. FTIR analysis showed that CAPE molecule was successfully encapsulated within the nanoparticles. Successful conjugation of angiopep-2 peptide and nanoparticles was verified with FT-IR, size and zeta potential results. Photon correlation spectroscopy analysis showed that size and zeta potential of the targeted PLGA nanoparticles were 176.4 nm and -10.8 mV, respectively. Biocompatibility and cytotoxic activity of free CAPE, CAPE loaded NPs and targeted NPs were comparatively investigated on L929 fibroblast and C6 glioma cell lines by MTT method. As a result of L929 fibroblast cell viability, both free drug and nanoparticles were biocompatible. IC50 values of the free CAPE molecule, CAPE loaded nanoparticles and angiopep-2 conjugated nanoparticles were 50, 45 and 21 µM, respectively. As a result of this master thesis, targeted nanoparticles showed greater anticancer activity than free CAPE molecule and CAPE loaded nanoparticles.
