Several studies reported differential involvement of two hemispheres in depression. One of the indicators of hemispheric asymmetry, handedness in humans and paw preference in rodents points to differential susceptibility to depression. The association between depression and cognitive impairments are also well documented. Thus we investigated the effect of paw preference on depressive like behavior and spatial learning as an indicator of cognitive function in male adult Wistar rats. Paw preference was determined in a task in which food deprived rats could obtain food from a feeding tube narrow enough to be accessible only using one paw. Rats were classified as right- or left-pawed if they preferred to use that paw in at least 70% of the 100 trials in the last two days of four day paw preference testing. Four days after last session of paw preference test rats were subjected to an open space swim test (OSST) which was conducted for 15 minutes daily on 3 consecutive days. In this model of depression, a decrease in distance moved and an increase in immobility on the third day compared to the first is interpreted as an indicator of depressive like behavior. Seven days after last OSST session rats were examined in Morris water maze (MWM) test with a hidden platform for 5 consecutive days (5 trials/day) to measure their performance in spatial learning. Results revealed that left-pawed rats decreased the distance moved and increased the immobility from first day to third more than right-pawed rats, indicating that left-pawed rats are more vulnerable to depressive-like behavior induced in OSST, although the results were not statistically significant. Then, rats were grouped as depressed or non-depressed depending on the change in the swimming distance from first to third session by taking median as a cutoff point. The MWM results revealed no significant effect of depressive-like behavior induced in OSST on the performance of finding the hidden platform. There was also no significant effect of paw preference on escape latency to find the hidden platform. The results indicate that paw preference may be a behavioral indicator of susceptibility to depressogenic treatment and future studies can focus on underlying molecular mechanisms.
