The link between inflammation and cancer has been weil-established and cytokines constitute the major component of this linkage, lnterleukin-17 (IL-17), one of these proinflammatory cytokines, plays an important role in autoimmunity, chronic inflammation and cancer. However, to date, studies examining the relation between tumour development and IL-17 expression are limited and controversial. The aim of this study was to examine the expression of IL-17 in human breast cancer and pancreatic cancer, as well as to explore the biological significance of IL-17 in tumour development. Macrophages in a proportion of breast cancer patients were found to be IL-17 positive whilst IL-17 positivity was determined in both macrophages and cancerous tissue in pancreatic cancer. In vitro studies in matrigel showed that IL-17 led to invasion in breast cancer cell but not in pancreatic cancer cell lines. Breast cancer cell invasion was matrix-metalloproteinase (MMP) dependent as evidenced by MMP inhibitors. Pancreatic immune staining further suggested that IL-17 expression correlated with better 4-year overall survival compared to IL-17 negative tumours. Thus this study has brought new insights into the context of IL-17 role in cancer biology and identified IL-17 as a potential prognostic marker in pancreatic cancer.
