Özet
42 ÖZET Doğu Akdeniz Bölgesindeki salep orkidelerinde, mikorizal fungusların teşhisi ve bu fungusların orkide tohumlarının çimlenmesi üzerine etkisinin araştırılması amacıyla yapılan bu çalışma 1993-1994 yılı boyunca yürütülmüştür. Yukarıdaki adı geçen bölgelerde 14 farklı orkide türü saptanmıştır. Bu 14 türün rizosfer topraklarından yapılan izolasyonlar sonucunda 2 Rhizoctonia solani izolatı ve 6 fungus cinsi elde edilmiştir. Orkide tohumlarının çimlenmesi ve gelişimi ile ilgili olarak Rhizoctonia solani olarak teşhis edilen izolat kullanılmıştır. Elde edilen diğer fungus cinslerinin Absidia sp., Fusarium spp., Macrophomina sp., bazı Pythiaceous funguslar ve Trichoderma sp. oldukları belirlenmiştir. Bu bölgelerde tesbit edilen Orchis coriophora, Orchis anatolica, Ophrys transchyrcana, Ophrys vernixia, Ophrys apifera, Ophrys umbilicata, Serapias vomaracea ve Himantogiossum af fine türleri tohumları ve Rhizoctonia solani, Absidia sp. ve Trichoderma sp. izolatları ile Knudson besin çözeltisi kullanarak petrilerde çimlendirme denemeleri kurulmuştur. Bu denemeler sonucunda Orchis coriophora ve Ophrys vernixia ' ya ait tohumlarda Rh-1'in varlığında çimlenme ve fide gelişimi gözlenirken, Rh-1'in bulunmadığı kontrol petrilerde protokorm oluşumu ve çimlenme gibi gelişmeler kaydedilmemiştir. Orchis anatolica tohumlanyla kurulan petri denemelerinde ise Rhizoctonia soiani sadece protokorm oluşumunu teşvik etmiş ve fide gelişimi gözlenmemiştir. Dolayısıyla Rh-1'in Orchis coriophora ve Ophrys vernixia üzerinde mikorizal etkisi kesinlik kazanırken Orchis anatolica' da etki protocorm oluşumuyla sınırlı kalmıştır.43 Fungus+Knudson çözeltisinin yanısıra ayrıca çimlendirme ortamı olarak kullanılan yulaf ortamında da Orchis coriophora + Rhizotonia solanı (Rh-1) ortaklığı çimlenmede başarılı sonuç vermiş ve sağlıklı orkide fidecikleri elde edilmiştir. Çimlendirme denemelerinin bir kısmı ayrıca orkide rizosferinden alınan bir miktar toprakla saksı denemesi kurularak gerçekleştirilmiştir. Saksı denemesinde belirli aralıklarla yapılan mikroskobik ölçümler sonucunda tohumlarda ne bir çimlenme ve ne de kayda değer başka herhangi bir değişiklik gözlenmemiştir.

44 SUMMARY The main goal of this study was to identify the mycorrhizal fungi on the root of salep orchids and to investigate the effects on the germination of orchid seeds. The project was carried out in the Eastern Mediterrenean region in 1993 and 1994. In this region, 14 different orchid species were obtained during surveys. 2 Rhizoctonia isolate, which belongs to R.solani and 6 other different fungus isolates isolated from the rhizosfer of these orchid species. Only one R.solani isolate called Rh-1 was used to test the effects of the fungus on the germination of the seeds and on the seedling developments. Fungus genara isolated from orchids were identified as Trichoderma sp., Fusarium spp., Macrophomina sp., Absidia sp. and some were as Pythiaceous fungi. The effects of R. solani isolates on the germination were tested in petri dishes containing Knudson nutrient solution. Seeds of Orchis coriophora and Ophrys vernixia germinated and showed some seedling development in the presence of Rh-1 while controls did not show any germination or protocorm development. Howewer the presence of R.solani stimulated the germination of Orchis anatolica but no seedling development observed. Mycorrhizal relationship between Orchis coriophora and Rhizoctonia solani was observed on oat-meal medium as well as Knudson medium. In this trials, orchid seeds germinated and healthy seedlings developed. Some of the germination tests were carried out using soil from the rhizospher of orchids in experimental pots. The seeds were periodically checked was seen in terms of germination or other developmental changes.
