The in vivo measurement of photosynthesis has been largely limited to fluorescence. which is hard to interpret being a very indirect measurement and to gas exchange in continuous light. which infers only about the overall efficiency of the leaf and does not give any quantitative data about the concentrations and absolute activities of photostynthetic complexes. In this study. a method. proposed for in vivo determination of the concentration and redox state of Photosystem I. one of the photoactive complexes of photosynthesis. has been corrected. Most of the complexes in the photosynthetic unit can be measured with exactly the same principles and similar equipment. This would make possible a diagnostic test system for nondestructive testing of photosynthetic stress The necessity of P700 measurement for such a system and the opportunities it promises have been briefly discussed. It has been concluded that the magnitude and exact site of damage on photosynthesis by environmental factors can be determined with te help of PS I measurement. For the quantitative justification of the measurements. the influence of the nonideal optical properties of the leaf on the measurements have been investigated by experimental means and it has been observed that the refraction and scattering phenomena in the leaf magnify the apparent absorption changes by a factor of ca. six. A method for the exact determination of active PS I centers, through measurement of the oxygen evolved by single turnover flash excitation has been developed. The elongation of the pathlenght of the light in the leaf up to about six times the thickness of the leaf has been found to be due to mostly the refractions in the lipid air interface at the spongy parenchima. This organisation of the leaf structure appears to be an intentional event, with the purpose of increasing the light harvesting efficiency of the leaf at the spectral region between the peaks of chlorophyll absorption.
