Dr. Zekiye SULUDERE Yrd. Doç. Dr. Hakkı SAYAR Agrotis segetum hemolenfinin sitotoksik aktivitesi in vitro şartlarda incelenmiştir. Hedef materyal olarak bakteri ve insan kan hücreleri kullanılmıştır. Hemolenf örnekleri hem immunize edilmemiş ve hem de hemosöle bakteri enjeksiyonu ile elde edilmiş immunize hemolenf olarak toplanmıştır, immunize edilmemiş hemolenf test edilen bakterilere karşı herhangi bir sitotoksik etki göstermemiştir. Sadece Bacillus subıilis ile immunize edilmiş hemolenf, aynı bakteri ile yayılmış Tryptone soy (TS) agar plaklarında sitotoksik bulunmuştur, immunize hemolenf, hemolenf sıvısı ve hemositler olarak ayrılmıştır. B. subtilis ile aşılanmış TS agar plaklara damlatılan bu iki fraksiyon bakteri gelişmesini inhibe etmiş ve bir inhibisyon zonu meydana getirmiştir, immunize hemolenfin insan kan hücrelerine karşı olan sitotoksisitesi Thoma lamı kullanılarak incelenmiştir. Bu testlerde eritrositlerin hemolize olduğu yerlerde plak şekilleri gözlenmiştir. Bu bölgelerde hemositler görülmüştür.

Prof. Dr. Hakkı SAYAR Cytotoxic activity of haemolymph withdrawn from Agrotis segetum larvae has been treated in vitro conditions. Bacteria and human blood cells were used as target material. Haemolymph samples were collected both non-immunized and immunized haemolymph obtained by injecting bacteria in haemocole. Non-immunized haemolymph did not show any cytotoxic activity against the tested bacteria. Immunized haemolymph by Bacillus subtilis was only found cytotoxic for same bacterial species which overlayed on Tryptone soy (TS) agar plaque. Immunized haemolymph was separated as haemolymph fluid and hemocytes. These two fractions which pooled in TS agar plate, seeded with Ö. subtilis, inhibited the growth of bacteria and formed an inhibition zone. Cytotoxicity of immunized haemolymph were treated against the human blood cells in jell using Thoma chamber. In these experiments, the patterns of plaques of hemolyzed erythrocytes were observed. Hemocytes could be seen in the pattern of plaques.
