Iloprost, a chemically stable analogue of prostacyclin, produced dose dependent anticonvulsive activity on pentylenetetrazol (PTZ) induced seizures following intracerebroventricular (icv) administration with an EDoo and (95 % confidence limits) 224.96 (100.43-504) ng/icv in rats. Since iloprost reduced locomotor activity only after higher doses and it was ineffective against strycn ine- induced seizures, specific mechanisms of action might be involved for the inhibitor effect on PTZ-induced seizures. Clonazepam pretreatment potentiated the anticonvulsive effect of iloprost. In this situation ED»o value and 95 % confidence limits of iloprost was 39.4 (23.89-65.01) ng/icv. Further, considerable less amount of iloprost injections into substansia nigra pars reticulata also produced anticonvulsive response against PTZ seizures. All these data implicated the role of GABAergic inhibitory system mediating anticonvulsive effect of iloprost in rats.
