Özet
ÖZET Lyon hipotezine göre dişi memeli hücrelerinde bulunan iki X kromozomundan biri erken embriyonik dönemde rastgele bir seçimle inaktif leşmektedirler (35). İnaktivasyona uğra yan X kromozomu hücre siklüsünün S fazında diğer kromozomla ra oranla daha geç bir dönemde replikasyona uğrar (19). Bu çalışmanın ilk aşamasında, geç replike olan X kromo zomunu göstermek için geliştirilmiş olan boyama yöntemlerinde üçü (Akridin Oranj, Harlequein Reverse, RBG) denenip, karşı laş tın Imışve hastalarda rutin olarak uygulanmak üzere RBG yöntemi seçilmiştir. Çalışmanın diğer bölümünde, anormal yapılı bir X kro mozomuna sahip şahıslarda inaktivasyon olayının tesadüfi ol maktan çıkıp genellikle tercihli olarak defektif X kromozomu nun inaktif leş tiği (17) bulgusundan yola çıkarak çeşitli mor folojik bozukluklar gösteren X kromozomlarına sahip 10 Turner Sendromu hastası kromozom inaktivasyonu yönünden incelen miştir. 46, X, i (Xq) kromozom konstitüsyonlu 4 hasta ve 45, X0/46,X,i (Xq) kromozom yapısına sahip 1 hastada incelenen metaf azlarda 547öor anında izokromozom olan X'in soluk boyanıp inaktive olduğu gözlenmiş tir.- 77 - 46,X(Xp-5 kro. ' konstitüsyonlu olguda %76 oranında defektif X soluk boyanırken, 46 X(Xm) kromozom yapısına sahip vakada Xm'in soluk olduğu sahaların oranı %37'dir. Ve 46, XX kromo zom yapısına sahip olgularda ise %73 oranında soluk X görül müştür. Hastalara ait hiçbir metafazda normal X kromozomu soluk olarak görülmemiştir. Elde edilen bulgular literatür le karşılaştırılmış ve uygunluk gösterdiği saptanmıştır.

SUMMARY According to the Lyon hypothesis, in female mamalian cells one of the two X chromosomes is inactivated during an early embryonic stage (35). In the S phase of the cell cycle the inactivated X chromosome replicates itself later than the other chromosomes (19). In the first part of this study three of the stainning methods (Acridin Orange, Alcali treated Giemsa, RBG) to show the late-replicating X have been compared and RBG is chosen the most suitable for routine patient analyses. In the other part of the study.; regarding to the finding that the st ructurally abnormal X within a cell is rather inactivated in a non-random fashion, the inactivation of the X chromosome have been analyzed in 10 Turner Syndrome patients having various types of st ructural abnormalities in their X chromosomes. In the metaphases of four patients having chromosome constution of 46, X, i (Xq) and one patients with a constitution of A5,X0/46 X,i (Xq) in72%of the cells the iso-X is shown to be inactivated and stainned differently without getting any bands. Xn the case- 79 - with 46, X (Xp-) the defective X is shown to be inactivated in 76% cells. Whereas in the case of 46, X (Xm), Xm stained in a uniform manner showing inactivation irjj^of the cells. In the cases with a karyotype 45, XO and 46, XX the percentage of unhanded X is found to be73ŞâHowever, it is worth to note that in none of the cases the normal X is shown to be inactivated. The findings of the study is compared with the previous work done in this field and found to be in great similarity.
