ÖZET Yüksek Lisans Tezi BAZI Solanum TÜRLERİNDE Verticillium dahliae'YE DAYANIKLILIK İLE FİTOALEKSİN OLUŞUMU ARASINDAKİ İLİŞKİNİN BELİRLENMESİ Mert SEVİN Ankara Üniversitesi Fen Bilimleri Enstitüsü Bahçe Bitkileri Anabilim Dalı Danışman: Prof. Dr. Şebnem ELLİALTIOĞLU Prof. Dr. A. Sülün ÜSTÜN Prof. Dr. İbrahim DEMİR Patlıcanda önemli zarar yapan Verticillium solgunluğu etmeni olan Verticillium dahliae fungusuna dayanıklılık özelliğinin, bitki dokularında sentezlenen fitoaleksin oluşumuyla bir ilgisinin olup olmadığını belirlemek; böylece geniş ölçekli bir patlıcan genotipi havuzunda bu hastalığa dayanıklılık özelliği bakımından bir tarama ve seçim yapmaya yönelik etkili bir yöntem geliştirmek amaçlanmıştır. Üç yabani patlıcan türü {Solanum sisymbriifolium, S.torvum, S.aethiopicum) ve üç yerli patlıcan çeşidine ait (Aydın Siyahı, Kemer, Topan 374) bir aylık meyvelere laboratuvar koşullarında V.dahliae elisitörü uygulanmıştır. 400 glukoz eşdeğeri olarak hazırlanan V. dahliae'nin otoklavlanmış spor süspansiyonu ile 48 ve 96 saat süreyle uyarım işlemine tabi tutulan örnekler, daha sonra derin dondurucuda saklanmışlardır. Doku ve süzüntüden ekstrakte edilen maddeler GLC'nde analiz edilmiştir. 'Genotip x uyan süresi x doku fazı' üçlü interaksiyonunun önemli bulunduğu çalışmada, fitoaleksin olarak lubimin belirlenmiş, en yüksek lubimin miktarı S. sisymbriifolium türünde 48 saatlik uyarı süresi sonunda saptanmıştır (9767 ug/ml) diğer iki yabani tür de lubimin birikimini yüksek düzeyde oluşturmuş, kültür çeşitlerindeki lubimin miktarı 48 saatlik uygulamada aynı istatistiksel grupta kalarak düşük değerler vermiştir. Fitoaleksin birikimi özelliği ile V.dahliae'ye dayanım arasında ilişki olabileceği yönünde bulgular bu çalışma ile daha da güç kazanmış, güvenilirliği tam bir test yöntemi olarak kullanılabilmesi için üzerinde uyarıcı cinsi ve dozu, bitki dokusu tipi gibi konularda çalışmalara devam edilmesi gerektiği görülmüştür. 2005, sayfa 61

Sülün ÜSTÜN Prof. Dr. İbrahim DEMİR It is intended to determine if there is a relation between the phytoalexin formation synthesized in plant tissues and the endurance characteristic of the Verticillium dahliae fungus, which is a factor of Verticillium paleness that causes important damages to eggplant; and by this way to develop an effective surveillance and selection method, in terms of the endurance characteristic against this illness, from the wide scale of eggplant genotype pool. In laboratory conditions, V. dahliae elicitor is practiced to the fruits of three kinds of foreign eggplant types {Solarium sisymbriifolium, S. torvum, S. aethiopicum) and three kinds of native eggplant types (Aydın Siyahı, Kemer, Topan 374). The samples which are stimulated for 48 and 96 hours with V dahliae'' % autoclaved spore suspension, that is prepared as the equivalence of 400 mgL_1 glucose, are stored in the deep frost machine. The substances, extracted from the tissue and the filtrate, are analyzed in GLC. In this research, 'genotype x warning time x tissue phase' trio-interaction is considered as important and fitoaleksin is determined as lubimin. after the 48 hours of warning time, the higher amount of lubimin amount is found in the S. sisymbriifolium type (9767 ng/ml), in the other two wild types the lubimin accumulation occurred in high amounts and lubimin amount in cultural samples resulted as low values by staying in the same group after the 48 hours treatment. The findings of a relation between the characteristics of phytoalexin accumulation and endurance of V. dahliae are supported with this research, and it is agreed that to use the reliability as a fully test method, the researches must be continued on the subjects like plant tissue types, stimulant type and dose. 2005, page 61
