Özet
SUMMARY Objective: Matrix mctalloproteinases (MMPs) and tissue inhibitors of matrix metalloproteinases (TIMPs) play an important role in fibrosis. In this study, we prepared an animal model of peritoneal fibrosis due to chemical peritonitis. We investigate the effects of MMP inhibitor, doxycycline on changes in peritoneal morphology, MMP and TIMP activities in peritoneal lavage fluids. Methods: 21 rats were used, divided into three groups (n- 7). Duration of the study was 14 days. In the first group, we administered intraperitoneally (i.p) chlorhexidine gluconate (CHX) to rats. In the second group, we administered i.p doxycycline in addition to CHX to rats. In the third group, we administered i.p saline to rats. At the end of the study; peritoneal lavage fluids, parietal and visceral peritoneum tissue samples were obtained. MMP-2, MMP- 9, TIMP-1, TIMP-2 and procollagen type I C-terminal peptid (PICP) levels in the peritoneal fluids were analyzed. Thickness of the peritoneum, amount of the immature collagen and the inflammation scores were measured in peritoneal tissue samples. Results: Parietal-visceral peritoneal thickness, parietal-visceral peritoneal inflamation scores and immature collagen levels in parietal peritoneum were compared and significant difference were observed between the groups (p<0.05). TIMP-1 and PICP levels in the peritoneal lavage fluids were compared and significant difference were observed between the groups (p<0.05). MMP-2 and TIMP-2 levels in the peritoneal lavage fluids were compared and no significant difference were observed between the groups (p>0.05). MMP-9 were not detected on gelatin zymography method in the peritoneal lavage fluids within the groups. Conclusions: In this experimental study, CHX solution leads to peritoneal injury in peritoneal tissue. Although insignificant difference is present statistically, MMP inhibition with doxycycline decrease the peritoneal injury due to CHX solution.
