Pentilentetrazolle oluşturulan in vivo ve invitro nöronal hasar modelleri sonrası kapsamlı amino asit profilinin değerlendirilmesi

Özet
Amaç: Bu çalışmada, in vivo ve in vitro modellerde pentilentetrazol (PTZ) ile oluşturulan nöronal hasar sonrası amino asit profilindeki değişimlerin incelenmesi amaçlanmıştır. Farklı biyolojik örnekler karşılaştırılarak nöronal hasar modellerinde amino asit değişikliklerine ilişkin bilgi elde edilmesi ve tedaviye yönelik yeni bir bakış açısı geliştirilmesi hedeflenmiştir. Materyal ve Metod: Çalışmada HT-22 fare hipokampal nöronal hücre hattı kullanıldı. PTZ ile oluşturulan nörotoksisite sonrası hücreler belirlenen zaman noktaları olan 12. saat ve 24. saatlerde toplanarak −80 °C'de saklandı. Kontrol grubuna herhangi bir işlem uygulanmadı. Hücre örnekleri analiz öncesinde dondur-çöz işlemi uygulanarak hazırlanıp ve santrifüj edilerek süpernatantlar elde edildi. İn vivo çalışmada ise deney hayvanlarına PTZ uygulanarak nöbet oluşumu sağlandı. Hayvanlardan 0.saat (PTZ uygulamasından sonraki ilk 10 dk.) içinde,12.saatte ve 24. saatte kan örnekleri alındı ve aynı zaman noktalarında kardiyak perfüzyon sonrası hipokampus dokuları çıkarıldı. Kan örnekleri santrifüj edilerek plazmaları ayrıldı ve -80°C'de saklandı. Doku örnekleri de alındıktan hemen sonra hızlı dondurma için -80 °C'de uygun koşullarda saklandı. Elde edilen hücre, plazma ve doku örnekleri homojenizasyon ve hazırlama işlemlerinin ardından LC-MS/MS cihazı ile amino asit analizleri yapılmıştır. Bulgular: Hücre kültürü, doku ve plazma örneklerinde PTZ uygulamasına bağlı olarak çeşitli amino asit düzeylerinde zamana bağlı değişiklikler gözlenmiştir. Hücre kültürü deneylerinde taurin düzeyleri 12. saatte kontrol grubuna göre anlamlı derecede azalmış (p<0.05), 24. saatte ise azalma eğilimi sürmesine rağmen istatistiksel anlamlılık göstermemiştir. Arjinin ve 2-aminobütirik asit düzeyleri 24. saatte kontrol grubuna kıyasla anlamlı derecede düşük bulunmuştur. Ayrıca 12. saatte 2-aminoadipik asit, etanolamin, O-fosforil etanolamin, 5-hidroksi lizin ve karnosin düzeylerinde belirgin azalma tespit edilmiştir. Doku örneklerinde metiyonin düzeyleri 12. saatte kontrol grubuna göre anlamlı şekilde yüksek bulunurken, beta-alanin düzeyleri 0. saatte anlamlı derecede düşük saptanmıştır. 2-aminobütirik asit 12. saatte kontrol grubuna göre düşük bulunmuş ancak bu değişim istatistiksel olarak anlamlı değildir. Sistin düzeyleri ise 24. saatte kontrol grubuna kıyasla anlamlı derecede artmıştır. Plazma örneklerinde taurin ve Gamma-Aminobütirik Asit (GABA) düzeyleri 12. saatte kontrol grubuna göre anlamlı derecede düşük bulunmuştur. Arjinin ve 2-aminobütirik asit düzeyleri 24. saatte 0. saat grubuna göre anlamlı şekilde artmıştır. Ayrıca glisin düzeyleri 24. saatte kontrol grubuna kıyasla anlamlı derecede yüksek tespit edilmiştir. Bunun yanında triptofan, 3-aminoizobütirik asit, valin, norvalin, glutamik asit, aspartik asit, treonin, serin, prolin, glutamin ve trans-4-hidroksiprolin düzeylerinde de zaman noktaları arasında anlamlı değişiklikler gözlenmiştir. Sonuç: Bu çalışmada, PTZ ile oluşturulan epileptik modelde hücre kültürü, doku ve plazma örneklerinde amino asit metabolizmasında zamana bağlı değişiklikler ortaya konmuştur. PTZ'nin neden olduğu epileptik stres, amino asit metabolizmasında hücre kültürü, doku ve plazmaya özgü, heterojen değişikliklere yol açmaktadır. Özellikle 2-AB, taurin, GABA ve metiyonin-sistin eksenindeki bozukluklar, amino asit dengesizliklerinin nöbet patofizyolojisinde kritik bir rol oynayabileceğini göstermektedir.Ayrıca glikofosfolipit metabolizmasında ve transsülfürasyon ve glutatyon metabolizmasında epileptik süreçte amino asit metabolizmasının önemli ölçüde etkilendiği görülmektedir. Bu bulgular, epilepsi patofizyolojisinde amino asit metabolizmasının önemli bir rol oynayabileceğini ve amino asitlerin yeni tedavi yaklaşımlarında rol oynayabileceklerini düşündürmektedir.

Aim: The aim of this study was to investigate the changes in the amino acid profile after pentylenetetrazole (PTZ)-induced neuronal damage in in vivo and in vitro models. By comparing different biological samples, it was aimed to obtain information about the molecular basis of the disease in neuronal models and to develop a new perspective on diagnosis and treatment. Materials and Methods: HT-22 mouse hippocampal neuronal cell line was used in the study. After PTZ-induced neuronal damage, cells were collected at the specified time points of 12 hours and 24 hours and stored at −80 °C. No action was taken on the control group. Cell samples were prepared by freeze-thaw process before analysis and centrifuged to obtain supernatants. In the in vivo study, a seizure model was created by applying PTZ to experimental animals. Blood samples were taken from the animals within 0 hours (first 10 minutes after PTZ application), 12 hours, and 24 hours, and hippocampus tissues were removed after cardiac perfusion at the same time points. Blood samples were centrifuged, plasma separated and stored at -80°C. Tissue samples were also stored at -80 °C under suitable conditions for rapid freezing immediately after collection. After homogenization and preparation of the obtained cell, plasma and tissue samples, amino acid analyzes were performed with the LC-MS/MS device. Results: Time-dependent changes in various amino acid levels were observed in cell culture, tissue, and plasma samples due to PTZ application. In cell culture experiments, taurine levels decreased significantly at 12 hours compared to the control group (p<0.05), and although the decreasing trend continued at 24 hours, it did not show statistical significance. Arginine and 2-aminobutyric acid levels were found to be significantly lower at 24 hours compared to the control group. In addition, a significant decrease in 2-aminoadipic acid, ethanolamine, O-phosphoryl ethanolamine, 5-hydroxy lysine and carnosine levels was detected at 12 hours. Methionine levels were found to be significantly higher in tissue samples at 12 hours compared to the control group, while beta-alanine levels were found to be significantly lower at 0 hours. 2-aminobutyric acid was found to be lower at 12 hours compared to the control group, but this change was not statistically significant. Cystine levels increased significantly at 24 hours compared to the control group. Taurine and Gamma-Aminobutyric Acid (GABA) levels in plasma samples were found to be significantly lower at 12 hours compared to the control group. Arginine and 2-aminobutyric acid levels increased significantly at 24 hours compared to the 0 hour group. In addition, glycine levels were found to be significantly higher at 24 hours compared to the control group. In addition, significant changes were observed in tryptophan, 3-aminoisobutyric acid, valine, norvaline, glutamic acid, aspartic acid, threonine, serine, proline, glutamine and trans-4-hydroxyproline levels between time points. Conclusion: In this study, time-dependent changes in amino acid metabolism were revealed in cell culture, tissue and plasma samples in the epileptic model created by PTZ. Epileptic stress caused by PTZ leads to heterogeneous changes in amino acid metabolism that are specific to cell culture, tissue and plasma. Disturbances in the 2-AB, taurine, GABA, and methionine-cystine axis, in particular, suggest that amino acid imbalances may play a critical role in seizure pathophysiology. It also appears that amino acid metabolism is significantly affected in glycophospholipid metabolism and in the epileptic process in transsulfuration and glutathione metabolism. These findings suggest that amino acid metabolism may play an important role in the pathophysiology of epilepsy and that amino acids may play a role in new treatment approaches.
