Özet
İnsan ömrünün uzaması ve bireysel aktivite ihtiyacının artması implant malzemelerine olan gereksinimi artırmaktadır. Bu kapsamda uzun süreler boyunca kullanılması gereken total kalça artroplastisi (THA), kalça hastalıklarının tedavisinde etkili bir yöntemdir. Ancak, uygulama sonrasında periprostetik kırık, aşınma ve implant gevşemesi gibi komplikasyonlar görülebilmektedir. THA implantına sahip hastaların yaş ortalamasının gerilemesi ve ortalama yaşam süresinin uzaması nedeniyle ikincil cerrahi gereksinimlerinin olası artışı beklenmektedir. Özellikle asetabular bileşenlerde meydana gelen kırılma ve aşınmaya bağlı başarısızlık, revizyon hastalarının ~%0,25-11'i arasında değişmektedir. Günümüzde, hastaların gereksinim ve ihtiyaçlarına, klinik çıktılara daha fazla odaklanan, kalitesi yüksek implant malzemelerine yönelik talep hızla artmaktadır. THA uygulamalarında kullanılan implant malzemesinin seçimi hem hasta hem de sağlık sistemi açısından uzun vadeli sonuçlar üzerinde kritik bir etkiye sahiptir. Kalça implantlarında en sık kullanılan seramik esaslı ürünlerden biri, itriyum oksit ile stabilize edilen tetragonal zirkonya partikülleri (YSZ) içeren alüminyum oksit (Al₂O₃) esaslı kompozitlerdir. Bu biyoseramikler, kalça implantlarının femoral baş ve asetabular yuva kısımlarında yaygın şekilde kullanılmaktadır. Vücutta kullanımı olan seramiklerin, sahip oldukları birçok üstün ve gelişmiş özelliğe rağmen düşük kırılma tokluğu değerleri ve zor işlenebilir olmaları nedeniyle tek başına kullanımları sınırlıdır. Bu sorunun çözülebilmesi için geliştirilen yöntemlerden biri, kompozit yapılar oluşturmaktır. Bu bağlamda kullanılacak implantın biyouyumluluk özelliklerini koruyarak yapıya eklenen ikincil fazlar sayesinde, biyoinert matrislerin kırılma tokluğu, yoğunluk, aşınma dayanımı ve işlenebilirlik gibi özellikleri iyileştirilebilmektedir. Yapılan araştırmalarda, zirkonya ile toklaştırılmış alümina (ZTA) matrisinde eklenen tabakalı yapının, kırılma tokluğunu artırmada, tek boyutlu malzeme veya partikül formundaki ilavelere göre daha etkili olduğu belirlenmiştir. Bu durum, hem biyouyumlu hem de kendiliğinden tabakalı bir yapıya sahip olan, katkılandığı kırılgan matrisin işlenebilirliğini iyileştirebilen ve çeşitli toklaştırma mekanizmalarıyla matrisin kırılma tokluğunu artırabilecek MAX fazlarının, ikincil faz (katkı) olarak kullanım potansiyelini araştırma fikrini gündeme getirmiştir. MAX fazları, Mn+1AXn (n = 1, 2, 3) genel formülü ile tanımlanmaktadır. M bir geçiş metalini, A çoğunlukla IIIA ve IVA (Al, Ga, vb.) grubu elementini ve X ise karbon (C) veya azotu (N) belirtmektedir. MAX fazları üçlü karbür ve nitrürlerin çok kristalli, nano boyutlu ve tabakalı yapılarıdır. MAX fazları yapılarında hem seramik hem de metalik faz ve bu fazların özelliklerini bulundurmaktadır. Seramik fazlar MAX'a yüksek sertlik, iyi oksidasyon direnci, yüksek sıcaklık kararlılığı gibi özellikleri sağlamaktadır. Metalik fazlar ise işlenebilirlik, termal şok direnci, yüksek termal ve elektriksel iletkenlik gibi özelliklere sahip olmasına neden olmaktadır. MAX fazları, bu özellikleri ile yüksek sıcaklık, sensör, koruyucu kaplama, ince film, düşük sürtünmeli yüzey, biyomedikal uygulamalarında ve metalurji, otomotiv, havacılık-uzay sanayiinde kullanılabilmektedir. Bu kapsamda yapılan doktora çalışması iki aşamada gerçekleştirilmiştir. İlk aşamayı, Tix+1SiCx (TSC) ve Tiy+1AlCy (TAC) MAX fazlarının sol-jel yöntemi ve ısıl işlemi ile sentezlenmesi oluşturmuştur. TSC ve TAC MAX fazlarının ham maddelerinin sol-jel yöntemi ile sentezlenmesine yönelik bir çalışma literatürde bulunmamaktadır. İlk aşama için sentezlenen MAX fazlarının yüksek saflıkta olmaları hedeflenmiş ve doktora çalışmalarının ikinci aşaması, sentezlenen MAX fazlarının biyoinert özellik gösteren Al2O3 ve ZTA (hacimce %20 ve %30 YSZ içeren Al2O3) matrislerine farklı oranlarda eklenmesi ile kompozit yapıların spark plasma sinterleme (SPS) ile üretilmesini kapsamaktadır. Sol-jel yöntemi ile sentezlenen MAX fazı ham maddelerinin ısıl işlem sonucunda yüksek verimde MAX fazı oluşturabilmesi için hem sol-jel hem de ısıl işlem aşamalarında optimizasyonlar gerçekleştirilmiştir. Kullanılan başlangıç ürünlerinin stokiyometrik oranları (Ti:Si ve Ti:Al, 2:1 ve 3:1 gibi), nihai üründe elde edilen karbürlü yapının oluşması için ekstra karbon ilavesi, hidroliz oranı ile ham maddenin fırın ortamında kurutulması, sol-jel kısmındaki optimizasyonları kapsarken ısıl işlem esnasında yan ürünlerin (oksitli bileşikler, TiO2 ve Al2O3 gibi) oluşumunu engellemesi oksijen depolama görevi gören MnO2'nin fırın ortamına ilavesi, bu sıcaklıkta tozların işleme tabi tutulması ısıl işlemdeki optimizasyonlara örnek olarak verilebilir. Bu aşamada elde edilen önemli çıktılar, TSC'nin yüksek sıcaklık (1350°C) ve MnO2 ortamında yüksek verimde oluşmasıyken TAC oluşumununn termodinamik değil kinetik olarak gerçekleşmesidir. Bu da daha düşük sıcaklıkta (1250-1300°C) daha fazla bekleme sürelerinde oluşması anlamına gelmektedir. Buna ek olarak, Ti2AlC düşük sıcaklıkta kararlı iken 1250°C üzerindeki ısıl işlemlerde Ti3AlC2'ye dönüşmektedir. Deneysel çalışmaların ikinci aşamasında Dr.Sinter 20.000 A kapasiteli 7.40 MK VII, SPS Syntex Inc. spark plazma sinterleme (SPS) sistemi ile 50 mm çaplı kompozitlerin üretimi gerçekleştirilmiştir. Üretilen ikili kompozitlerin, SPS yöntemi ile yüksek yoğunluklarda üretilmesi hedeflenmiştir. Alümina ve ZTA matrisleri içerisine farklı hacim oranlarında katkılandırılan MAX fazlarının relatif yoğunlukları hesaplanmış ve ZTA (70-30) matrisinin daha yüksek densifikasyon değerlerine ulaştığı belirlenmiştir. Ti3SiC2 ve Ti3AlC2 MAX fazları aynı oranlarda (hacimce %0,5, 1, 3, 5) ve aynı SPS parametrelerinde (1350°C-60 MPa- 5dk) ZTA (70-30) matrisine eklenmiştir. Hacimce eklenen %3 MAX fazları her iki sistem için de en iyi sonuçları vermiş olup işlenebilirlik çalışmaları yalnızca bu kompozitler için gerçekleştirilmiştir. Longer human lifespan and the need for increased individual activity increase the need for implant materials throughout life. Total hip arthroplasty (THA) is an effective method for the treatment of hip diseases. However, complications such as periprosthetic fracture, wear and implant loosening may occur after THA. The failure rate of primary surgical interventions after THA is approximately 12% in a 10-year follow-up. As the average age of patients undergoing, THA is decreasing and life expectancy is increasing, the need for revision surgery is expected to increase gradually. Failure due to fracture and wear, especially in acetabular components, is seen in a rate ranging from 0.25% to 10.9% of revision patients. Today, the demand for high quality implant materials that are more focused on patient needs and clinical outcomes is rapidly increasing. The choice of implant material used in THA applications has a critical impact on long-term outcomes for both the patient and the healthcare system. One of the most commonly used ceramic-based products in THA applications is aluminum oxide (Al₂O₃)-based composites containing tetragonal zirconia particles. Zirconium oxide (ZrO₂) increases the fracture toughness of alumina and is very important in implant designs. It undergoes a reversible phase transformation from tetragonal to monoclinic, resulting in 4-5% volume expansion and 14-15% shear stresses. This transformation is detrimental to implant surfaces in contact with body fluids and leads to microcracks and surface roughening. Stabilizing oxides such as MgO, CaO and Y₂O₃ prevent phase transformation at room temperature, with Y₂O₃ being the most effective due to its positive effect on toughness. Alumina (3-YSZ) combined with yttria-stabilized zirconia (YSZ) outperforms monolithic alumina and zirconia in many structural applications. Despite their many superior and advanced properties, ceramics used in the body have some limitations when used alone due to their low fracture toughness values and difficult machinability. Low fracture toughness can lead to slow progressive crack formation in the long term and the implant may fail to fulfill its function. One of the methods developed to solve this problem is the creation of composite structures. By adding secondary phases to the structure while maintaining biocompatibility, properties such as fracture toughness, density, wear resistance and machinability of bioinert matrices can be improved. MAX phases are defined by the general formula Mn+1AXn (n = 1, 2, 3). M stands for a transition metal, A for a group element, mostly IIIA and IVA (Al, Ga, etc.) and X for carbon (C) or nitrogen (N). MAX phases are polycrystalline, nano-sized and layered structures of ternary carbides and nitrides. MAX phases contain both ceramic and metallic phases and their properties. Ceramic phases provide MAX with properties such as high hardness, good oxidation resistance, high temperature stability. The metallic phases, on the other hand, give MAX properties such as machinability, thermal shock resistance, high thermal and electrical conductivity. With these properties, MAX phases can be used in high temperature, sensor, protective coatings, thin films, low friction surfaces and biomedical applications and metallurgy, automotive, aerospace industries. The layered structure in zirconia toughened alumina (ZTA) matrix was found to be more effective in increasing fracture toughness than additions in the form of one-dimensional material or particles. This led to the idea of investigating the potential of using MAX phases, which are both biocompatible and self-layered, can improve the processability of the brittle matrix to which they are doped and can increase the fracture toughness of the matrix through various toughening mechanisms, as a secondary phase (additive). The PhD thesis was realized in two phases. The first stage was the synthesis of Tix+1SiCx and Tiy+1AlCy MAX phases by sol-gel method and calcination heat treatment. There is no study in the literature on the synthesis of Tix+1SiCx and Tiy+1AlCy MAX phases by sol-gel method. In the study, which aims to synthesize Ti3SiC2, Ti3AlC2 and Ti2AlC, experiments were first carried out for the synthesis of the MAX phase containing titanium and silicon. While the titanium source required to form the MAX phase was aimed to be provided by titanium (IV) butoxide, different chemicals [(GPTMS(GLYMO)=3-glycidyloxypropyltrimethoxysilane (CH3O)3-Si-CH2CH2CH2CH2OCH2C2H3O), and triethoxyoctylsilane=(C2H5O)3Si-CH2(CH2)6CH3)] were tried as silicon sources. The carbide of the carbide compound to be formed was obtained from the carbons in the starting materials (alkoxides). Hydrolysis of the solutions was carried out in a base medium and 1.0 molar tetrapropyl ammonium hydroxide (Pr4NOH(aq)) was used. It is considered that the oxides bound to titanium and silicon are removed with the carbons present in the hydrolysis product. In other words, it is suggested that some of the required carbons are met by the carbons present in the product and some of them are met by the carbon added from outside. Although there is enough carbon (butoxy, methoxy, glycidyloxypropyl) in the hydrolysis product to prepare carbide, it is believed that some of it evaporates and some of it is removed from the environment due to the high temperature cracking process. Stoichiometric ratios of 1:1, 2:1 and 3:1 were studied during the sol-gel preparation of the raw materials of MAX phases. Among these ratios, the 3:1 ratio was the most effective to form the Ti3SiC2 phase, while the 2:1 ratio gave the best result to synthesize Ti2AlC and Ti3AlC2. The solvent of the solution obtained after the hydrolysis step and the ethanol released were removed by vacuum evaporator under reduced pressure. The synthesized solid-gel like products were first ground in an agate mortar and powders with fine particles were obtained. Then, they were heat treated at 1350°C under argon at a heating rate of 2 oC/min and kept at this temperature for 60 min and cooled under furnace control. Due to the oxygen content of sol-gel products, oxygenated compounds (TiO2, SiO2 and Al2O3) were formed as well as the MAX phase as a result of heat treatment. In order to increase the purity of MAX phases, various acid solution purification processes were performed and the desired value (98%) could not be obtained. Therefore, compounds with the ability to absorb ambient oxygen in the heat treatment stage following the sol-gel process were investigated. Compounds such as CeO2 and MnO2 were reported to have oxygen absorbing properties. During the heat treatment experiments, MAX phases and MnO2 were placed in the furnace environment. The removal of oxygenated compounds was determined by XRD analysis and X'pert HighScore software. For the first stage, the high purity of the synthesized MAX phases was targeted and the second stage of the PhD studies involved the production of composite structures by adding the synthesized MAX phases at different ratios to Al2O3 and ZTA (Al2O3 containing 20% and 30% YSZ by volume) matrices with bioinert properties. In the second stage of the experimental studies, 50 mm diameter composites were produced with spark plasma sintering (SPS) system (7.40 MK VII, SPS Syntex Inc.). It was aimed to produce the produced binary composites at high densities by SPS method. The densification behavior of the composites was determined, phase analysis and microstructure characterization were performed, Vickers hardness, fracture toughness, flexural strength were measured and wear behavior was determined. After the structure-property-performance relationship was established through systematic characterization activities, composites with the best combination of densification and mechanical properties were identified for each matrix. The relative densities of MAX phases doped into alumina and ZTA matrices at different volume ratios were calculated and it was determined that the ZTA (70-30) matrix reached higher densification values. Ti3SiC2 and Ti3AlC2 MAX phases were added to the ZTA (70-30) matrix at the same ratios and at the same SPS parameters (1350°C-60 MPa-5 min). 97ZTA3TSC binary composite showed the best mechanical properties.
